二甲基亚砜(DMSO)对人肝癌 HepG2细胞的诱导分化作用的实验研究

     

摘要

Objective:To investigate the inducing differentiation effect of DMSO on human hepatocellular carcino-ma HepG2 cells. Methods:HepG2 cells were cultured in vitro and treated with DMSO. Effect of cell proliferation ac-tivity was measured with MTT assay. To observe the changes of cell morphology with inverted microscope and Giemsa staining,the alpha fetoprotein(AFP)and albumin(ALB)secretion were tested with radioimmunoassay. Gamma glu-tamyl transpeptidase(γ - GT)and alkaline phosphatase( ALP)activity were tested with enzymatic reaction. Re-sults:DMSO can significantly inhibit HepG2 cell proliferation after 72h(P ﹤ 0. 05),the morphology of the cells were differented to mature cell,AFP secretion and γ - GT enzyme activity were significantly decreased(P ﹤ 0. 05),ALP activity and ALB content were significantly increased(P ﹤ 0. 05). Conclusion:DMSO can induce human hepatocellu-lar carcinoma HepG2 to normal cell differentiation and inhibit cell proliferation.%目的:探讨二甲基亚砜(DMSO)对人肝癌 HepG2细胞的诱导分化作用。方法:体外培养人肝癌HepG2细胞,用 DMSO 干预细胞,MTT 法检测药物对细胞增殖活力的影响;倒置相差显微镜和瑞氏-姬姆萨染色观察细胞形态学的变化;放射免疫法检测细胞甲胎蛋白(AFP)和清蛋白(ALB)的分泌量;酶促反应试剂盒检测细胞中γ-谷胺酰转肽酶(γ- GT)、碱性磷酸酶(ALP)的活性。结果:用 DMSO 处理 HepG2细胞72h后,能显著抑制细胞的增殖(P ﹤0.05);细胞的形态向成熟细胞分化;细胞 AFP 的分泌量和γ- GT 酶的活性明显降低,ALP 活性和 ALB 含量则显著升高(P ﹤0.05)。结论:DMSO 具有诱导人肝癌 HepG2向正常细胞分化的作用并抑制细胞增殖。
