miRNA-302a在子宫内膜癌组织中的表达及其与临床病理特征的关系

Objective:To investigate the expression of the miR-302a in endometrial cancer tissue and to study its correlation with different clinical pathological factors.Methods:The qRT-PCR was used to detect the miR-302a expression in 34 cases tissue specimens of endometrial cancer and 30 cases of normal endometrial tissue,which are collected,and analyze the relationship between the expression level of miR-302a and the clinical pathological factors.Results:That the expression level of miR-302a in endometrial cancer tissue was lower than that in normal endometrial tissue which indicated the statistically significant difference(P=0.049,<0.05) via the Quantitative real-time PCR.Moreover,the expression level of miR-302a was statistically different in well differentiated,moderately differentiated,poorly differentiated endometrial cancer tissue(P=0.017,<0.05).miR-302a expressesd statistically differently in depth of infiltration of endometrial cancer tissue(P=0.012,<0.05).However,miRNA-302a expression was not significantly associated with the other clinical pathological factors.Conclusion:Compared with normal endometrial tissues,the expression level of miRNA-302a was significantly lower in endometrial carcinoma tissues.And also,the expression level of miR-302a was extremely related with well differentiated,moderately differentiated,poorly differentiated endometrial cancer tissue and the depth of infiltration of endometrial cancer tissue.It suggests that miRNA-302a may play a role in inhibiting cancer gene.%目的:检测miRNA-302a在子宫内膜癌(endometrial cancer,EC)组织中的表达,研究其与临床病理特征之间的关系.方法:应用实时荧光定量PCR(Real-time PCR)方法检测34例子宫内膜癌组织及30例正常子宫内膜组织中miRNA-302a的相对表达量,并分析miRNA-302a在不同临床病理资料的子宫内膜癌组织中的表达的差异性.结果:miRNA-302a在子宫内膜癌组织中的表达水平低于在正常子宫内膜组织中的表达水平(P=0.049,<0.05),在不同的组织学分化程度(高、中、低分化)的子宫内膜癌组织中,miRNA-302a的表达存在统计学差异(P=0.017,<0.05);在不同的肌层浸润深度(深度≤1/2、深度>1/2)的子宫内膜癌组织中miRNA-302a的表达具有统计学差异(P=0.012,<0.05);而miRNA-302a在子宫内膜癌组织中的表达水平与病理手术分期、淋巴转移及脉管浸润等临床病理资料之间无明显统计学关系(P>0.05).结论:与正常子宫内膜组织相比,miRNA-302a在子宫内膜癌组织中的明显低表达,并且与组织学分化程度(高、中、低分化)、肌层浸润深度(深度≤1/2、深度>1/2)有关提示其可能发挥抑癌基因的作用.
