目的:通过对相关数据库数据分析,探究喉癌组织中SP100基因表达情况和生物学意义,以及对患者生存和预后的影响和可能存在的靶向治疗的意义.方法:对NCBI中GEO数据库中的数据集数据进行表达量分析、生存分析和相关性分析.结果:研究分析表明,与正常癌旁组织相比,SP100基因在喉癌组织中存在明显的低表达(P=0.003 9),而在各癌症进程分组间其表达差异没有统计学意义;SP100基因的表达与PLAU和MMP1基因的表达均存在明显的负相关(PLAU,P=0.019 6,r=-0.516 8;MMP1,P=0.004 4, r=-0.608 8);SP100的低表达会导致喉癌患者的无复发生存率明显降低(GSE27020,P=0.002 4;GSE25727,P=0.017 9).结论:通过分析表明,SP100基因在喉癌组织中表达下调,提示该基因在恶性肿瘤的发生发展中起到了一定作用,可能是喉癌一个新的肿瘤抑制基因,并且对于喉癌治疗的特异性用药靶点的选用和预后检测都具有重要意义.%Objective:To explore the expression and biological significance of SP100 based on relevant database data analysis,the impact on the survival and prognosis of laryngeal carcinoma patients and the significance of targeted therapy.Methods:To sets of GEO in the NCBI database,to perform express analysis,survival analysis and correlation analysis.Results:Analysis showed that the expression of SP100 gene in laryngeal carcinoma was significantly lower than that in normal pericancerous tissue(P=0.003 9),but there was no significant difference between the cancer progression groups.There was a significant negative correlation between the expression of SP100 and PLAU and MMP1 (PLAU,P=0.019 6,r=-0.516 8;MMP1,P=0.004 4,r=-0.608 8).Low expression of SP100 results in a significant reduction in recurrence-free survival in patients with laryngeal cancer (GSE27020,P=0.002 4;GSE25727,P=0.017 9).Conclusion:Analysis showed that the expression of SP100 gene in laryngeal carcinoma was down-regulated,suggesting that this gene played a role in the development and progression of malignant tumor.It may be a new tumor suppressor gene in laryngeal carcinoma,and has a great significance for the selection of specific drug target for laryngeal cancer treatment and prognosis detection.
