固相吸附核酸提取法及其在丙型肝炎病毒RNA提取中的应用

         

摘要

Hepatitis C virus (HCV) is a type of important microorganism leading to iver cancer and other liver diseases. Therefore,detecting HCV RNA content in clinical specimens is known as the "gold standard" to diagnose HCV infection. However, in the process of RNA extraction, the widespread presence of RNA enzyme can affect the effectiveness of HCV RNA extraction. A solid-phase adsorption method can extract nucleic acid from clinical specimens,which has the advantages of simple to operate, good quality, high yield and using materials without poisoning effect on the human body. Now, this method has been widely applied in clinical detection of HCV RNA. This article will summarize the mechanism of solid phase nucleic acid extraction and its development in hepatitis C virus RNA extraction.%丙型肝炎病毒(HCV)是引起肝硬化、肝癌等肝脏疾病的重要病原微生物.目前检测临床标本的HCV RNA含量是判断HCV感染的"金标准".但在RNA提取过程中,广泛存在的RNA酶对HCV RNA的有效提取影响较大.固相吸附法提取临床标本中的核酸成分操作简便,核酸产量高、质量好,采用对人体无毒害作用的材料,目前该方法已在临床HCV RNA检测中广泛应用.本文将对固相吸附核酸提取法的原理及其在HCV RNA提取中的研究进展进行综述.
