Objective To explore the expression of Bcl-6 mRNA in the Tfh cells of HIV/AIDS patients and the relationship between Bcl-6 mRNA and the progression of HIV/AIDS.Methods This experiment chose 60 patients who were confirmed by HIV antibody test positive,during May 2014 to November from AIDS Research Institute in the First Affiliated Hospi-tal,Henan College of Traditional Chinese Medicine.According to the amount of CD4+T cells,the patients with HIV/AIDS were divided into A(CD4<200 cells/μl),B(200 cells/μl500 cells/μl)three groups.The Tfh cells expressed CD4+ CXCR5 + ICOS+ and lymphocyte subpopulation including T,B and NK cells were detected by flow cytometry (FCM).The expression of Bcl-6 mRNA levels were detected by reverse transcription-polymerase chain reaction (RT-PCR).The association between Tfh cells and Bcl-6 mRNA,B cells were analyzed.Results The expression of Bcl-6 mRNA from patients of A,B and C three groups was increased than in healthy individuals (2.94±0.91,2.27±0.62,2.15± 0.351,P <0.05).The percent of Tfh cells from group A patients was higher than group C patients (5.88±3.01 vs 1.26± 0.87,P =0.032)and also the healthy control group (5.88±3.01 vs 0.78±0.42,P =0.004).The percentage of NK cells and B cells in HIV/AIDS patients was no statistically significant.Correlation analysis showed that there was positive corre-lation between the proportion of Tfh cells and Bcl-6 mRNA (r=0.799,P =0.000).And Tfh cells were observed no relation-ship with B cells (r=-0.083,P =0.657).Conclusion The changes of Bcl-6 mRNA and Tfh cells may be related to the progression of HIV/AIDS.%目的：探讨 Bcl-6 mRNA 在 HIV／AIDS 患者滤泡辅助性 T 细胞（Tfh）中的表达及其与疾病进展的关系。方法筛选2014年5月～11月期间河南中医学院第一附属医院艾滋病临床研究中心经 HIV 抗体确证实验阳性者60例，根据CD4＋ T 细胞数将 HIV／AIDS 患者分为 A（CD4＜200个／μl），B（200个／μl＜CD4＜500个／μl）和 C（CD4＞500个／μl）三组，同期选取20例健康对照组，采用流式细胞术（FCM）检测淋巴细胞亚群（T，B，NK 细胞）和 Tfh 细胞（CD4＋ CXCR5＋ICOS＋）的百分比。实时荧光定量 PCR 技术检测 Bcl-6 mRNA 的表达，并分析 Tfh 细胞与其转录因子 Bcl-6 mRNA 的相关性，Tfh 细胞与 B 细胞的相关性。结果A，B 和 C 三组 Bcl-6 mRNA 表达均高于健康对照组（2.94±0.91，2.27±0.62，2.15±0.351，P 值均＜0.05）；A 组 Tfh 细胞大于 C 组（5.88±3.01 vs 1.26±0.87，P ＝0.032）和健康对照组（5.88±3.01 vs 0.78±0.42，P ＝0.004）；四组 NK 细胞和 B 细胞比较差异无统计学意义（P ＝0.364，0.571）；Tfh 细胞与 Bcl-6 mRNA正相关（r＝0.799，P ＝0.000），Tfh 细胞与 B 细胞无关联（r ＝－0.083，P ＝0.657）。结论Bcl-6 mRNA 和 Tfh 细胞的变化可能与 HIV／AIDS 疾病进展有关。
