类风湿性关节炎患者PBMC表达免疫球蛋白样转录子研究

摘要

目的 分析免疫球蛋白样转录子(ILTs)在类风湿性关节炎(rheumatoid arthritis,RA)患者外周血单个核细胞(PBMC)上的表达,探讨其与RA发病间的关联.方法 应用流式细胞术检测40例RA患者、36例健康PBMC上ILTs的表达.结果 RA患者CD3+T细胞上表达的ILT2平均相对荧光强度(MRFI)及ILT2阳性表达的CD56+ NK细胞数量显著高于健康对照组(P均＜0.05);RA中疾病活动度组的ILT-2阳性表达的NK淋巴细胞(％)高于低疾病活动度组(P＜0.05);根据Spearman等级相关分析,各类白细胞上ILTs表达的MRFI两两之间呈正相关(P均＜0.01),但与RA疾病活动度DAS28得分、病程不相关(P均＞0.05).结论 ILT2在T细胞、NK细胞的表达异常升高可能与RA发病有关.%Objective To study the expression of immunoglobulin - like transcript 2 ( ILT2) in CD3 + , CD19 + and CD56 + lympho-mas, and ILT4 in CD14+ cells from patients with Rheumatoid Arthritis ( RA). Methods Forty RA patients and 36 healthy controls were studied. ILTs expression was analyzed by flow cytometry in peripheral blood mononuclear cells ( PBMC). Results We found a significant increased expression of ILT2 by lymphocytes in RA patients. When the expression of this receptor was assessed in cell subsets, significantly higher MRFI levels were detected in CD3 +T lymphocytes ( P < 0.05) , and a significantly higher percentage of ILT2 + cells were detected in CD56 + NK from RA patients compared with healthy controls(P < 0. 05) , and they were well related (P < 0. 05) , however, but they were not related to DAS 28 score, RA stages. And also a significantly higher percentage of ILT2 + cells were detected in CD56 +NK from RA group of 3. 2 < DAS 28 < 5. 1 compared with RA group of DAS 28 < 3. 2 (P < 0. 05 ). Conclusion The abnormal expression of ILT2 detected in RA suggestes that they mainly participate in the pathogenesis of this condition.
