肌肉瘤小鼠模型建立及其血管内皮生长因子C、D的表达

     

摘要

To establish rhabdomyosarcoma model for researching the mechanism of lymphatic and hematogenous metastasis. To provide theoretical basis for clinical myosarcoma treatment. Methods Getting 30 sanitation Kunming mousse with the SI 80 cancerous ascites in mice, S - 180 Sarcoma cell lines in vitro, cells in culture after the bottle is full of single -layer to collect cells, made of 5 X 107/ml cell suspension. Use of stable 4-5 cell, planted in mice hip muscle in the same parts, build muscle flbrosarcoma tumor in situ cultivation models, observing the growth of tumors. To put the mouses to death in different time after lweek. Cut the tumors and detect volume and weight of tumors. Open chest and abdomen to observe condition of neoplasm metastasis and the realation of tumor vessel and the growth of lymphatic and tumor metastatic. Get the tissue of tumor for embedding and slicing. Use the method of immunohistochemisty and Western blotting to detect the expression of VEGF - C and VEGF - D. Results The amount of lymphatic vessel have significant increasing during growth of tumors in centre and marginal zone of rhabdomyosarcoma tissue. It shows that rhabdomyosarcoma tissue have lymphatic hyperplasia and neogenesis. It may have obvious correlation with the induction of VEGF - C and VEGF - D. Conclusion Rhabdomyosarcoma tissue (lymphatic vessels) increased expression of VEGF - c and VEGF - D, VEGF - C and VEGF - D is an important effect of Lymphangiogenic factors, so as to promote lymphatic metastasis of rhabdomyosarcoma.%目的 建立横纹肌肉瘤模型,研究肌肉瘤淋巴道转移机制,为临床肌肉瘤治疗提供理论依据.方法 取清洁级昆明系小鼠30只,用S180小鼠癌性腹腔积液,体外培养S-180肉瘤细胞系,待细胞在培养瓶内长满单层后,收集细胞,制成5×107/ml的细胞悬液.利用稳定4~5代的瘤细胞,在小鼠臀部相同部位肌层种植,建立肌纤维肉瘤的原位瘤种植模型,观察肿瘤的生长情况.1周后处死小鼠,切取瘤体,检测肿瘤体积、重量及开胸、腹观察肿瘤转移情况及不同时期肿瘤血管、淋巴管生长与肿瘤生长转移的关系.取肿瘤组织包埋制备切片,应用免疫组化、Western blotting等方法检测血管内皮生长因子C、D(VEGF-C、VEGF-D)蛋白质表达.结果 横纹肌肉瘤组织中心区和边缘区均存在淋巴管,数量随肿瘤的生长明显增加,这预示着横纹肌肉瘤组织中存在淋巴管的增生和新生,其可能与VEGF-C和VEGF-D的诱导密切相关.结论 横纹肌肉瘤组织(淋巴管)中VEGF-C和VEGF-D的表达增高,VEGF-C和VEGF-D是重要的促淋巴管形成因子,从而促进横纹肌肉瘤的淋巴道转移.
