紫草素对人胃癌细胞SGC-7901侵袭和迁移能力影响

     

摘要

Objective To investigate the effect of shikonin on invasion and metastasis of human gastric carcinoma SGC -7901 cells. Methods MTT assay was used to examine the effect of shikonin on proliferation of SGC - 7901 cells. Transwell chamber was performed to determine the invasion and migratory capacity of the cells. MMP -2, MMP -7 mRNA expression levels of SGC -7901 cells treated for 24h were measured by real - time fluorescence quantitative polymerase chain reaction ( RFQ - PCR ) . Results Shikonin can inhibit the proliferation of SGC - 7901 cells with obvious dose - dependent and time - dependent manner before 2μg/ml. Shikonin significantly inhibited invasion and migratory capacity of SGC -7901 cells treated for 48h. Relative MMP -2 and MMP -7 mRNA expression of SGC -7901 cells treated for 24h was significantly lower than that in the corresponding negative control group. Conclusion Shikonin inhibits the migration and invasion capacity of SGC -7901 cells in vitro and the mechanism may be associated with the down - regulated expression of MMP -2 and MMP -7mRNA.%目的 观察紫草素对人胃癌细胞SGC-7901增殖、侵袭、迁移能力的影响.方法 MTT法检测细胞增殖抑制作用,Transwell小室侵袭迁移实验检测细胞侵袭、迁移能力.实时荧光定量PCR测定加入不同浓度紫草素24h的SGC-7901细胞的MMP-2、MMP-7 mRNA的表达水平.结果 紫草素可以抑制人胃癌细胞的增殖,在2μg/ml之前其抑制效应呈浓度-时间依赖性;紫草素作用胃癌细胞株SGC-7901细胞48h后SGC-7901细胞的侵袭、迁移能力降低;紫草素作用胃癌细胞株SGC-7901细胞24h后,紫草素组的MMP-2的相对表达量均显著低于阴性对照组的相对表达量.结论经紫草素作用后SGC-7901细胞的侵袭、迁移能力降低,其机制可能与下调MMP-2、MMP-7 mRNA的表达有关.
