散发性乳腺癌组织中磷酸化BRCA1的表达及临床意义

     

摘要

目的 研究散发性乳腺癌组织中磷酸化BRCA1(p-BRCAl)的表达情况,并探讨其临床意义.方法 242例散发浸润性乳腺癌石蜡标本构建组织芯片,利用免疫组化方法检测p-BRCA1的表达情况,分析与临床病理参数之间的关系.结果 p-BRCA1在乳腺癌组织中阳性表达率为61.6% (149/242).p-BRCA1阴性者平均年龄低于p-BRCA1阳性者,差异有统计学意义(P<0.05).p-BRCA1表达率在绝经前、高组织学分级、大肿瘤直径、三阴性乳腺癌的病例中显著降低(P<0.05),在不同病理类型、区域淋巴结、TNM分期、ER、PR、HER-2状态之间的差异无统计学意义(P>0.05).结论 利用免疫组化方法检测p-BRCA1的表达状态,将有助于判断散发性乳腺癌的生物学行为并制定个体化的治疗方案.%Objective To detect protein expression of phosphorylated BRCA1 ( p - BRCA1) in sporadic breast cancer and to reveal the relevant clinical significance. Methods Totally 242 cases of sporadic invasive breast cancer tissue were collected to construct tissue microarrays. Immunohistochemistry was used to detect protein expression of p - BRCA1. The correlation between protein expression and clinicopathological parameters was explored. Results The positive rate of p - BRCA1 in breast cancer was 61. 6% ( 149/242). Average age of p - BRCA1 - negative patients was less than that of p - BRCA1 - positive ones( P < 0.05 ) . The protein expression was significantly lower in the cases with premenopausal status, high histological grade, large tumor size and triple -negative breast cancer ( P <0. 05 ) . No statistically significant difference was found in p - BRCA1 expression according to different pathological type, regional lymph node metastasis, TNM stage,ER, PR and HER -2 status. Conclusion Detection of p - BRCA1 protein expression by immunohistochemistry may help to determine the biological behaviour of the sporadic breast cancer and to develop individualized treatment programs.
