抗草甘膦转基因大豆对雄鼠睾丸的影响

     

摘要

以雄性昆明小鼠为动物模型,分别以抗草甘膦转基因大豆和非转基因大豆饲料喂食,于30、90 d后,取其睾丸.采用流式细胞术检测睾丸组织中各生殖周期细胞比例、Hoechst33258染色观察睾丸细胞凋亡、组织化学染色技术观察睾丸组织病理学变化.研究抗草甘膦转基因大豆对雄鼠睾丸的影响.结果显示,30、90 d实验组和对照组小鼠睾丸精细胞单倍体、2倍体、4倍体比例均正常且趋于稳定,单倍体比例占主导地位,且实验组与对照组相比无统计学差异.在荧光显微镜下实验组睾丸细胞的细胞核呈正常的蓝色,与对照组相比,未发现明显的细胞核呈致密浓染,或呈碎块状致密浓染,颜色有些发白的凋亡细胞.组织病理观察也未发现两组小鼠的睾丸出现明显病理损伤.30 d喂食抗草甘膦转基因大豆急性毒理研究和90 d喂食抗草甘膦转基因大豆亚慢性毒理研究对雄鼠睾丸均无显著影响.%MaleKunmingmicemodelsexposuretoglyphosate-resistanttransgenicsoybeanandNon-GMO soybean were established and used to evaluate the impacts of glyphosate-resistant transgenic soybean on the testis.The mice were fed for 30 and 90 day with glyphosate-resistant transgenic soybean and Non-GMO soybean,respectively,and then the mice testes were collected.Changes of testicular tissue cell re-productive cycles were detected by flow cytometry,and testicular tissue cell apoptosis were assessed by Hoechst 33258 dye.The pathological changes of mice testes were examined by paraffin sections.The re-sults showed that the percentages of testis cell populations (haploid,diploid,tetraploid)was no difference between the experimental group and control group (P>0.05 ),and the apoptosis of white testicular cell, nuclear thick dense or fragmental thick dense was not observed.The pathological changes of testicular tis-sues were also not observed in paraffin sections.There was no significant difference in male mice testicu-lar tissue changes between transgenic soybean and Non-GMO soybean.The acute toxicology research by 30 day feeding glyphosate-resistant transgenic soybean and the chronic toxicology research by 90 day feed-ing glyphosate-resistant transgenic soybeans were no difference.
