新生期母子分离对大鼠2型糖尿病进程的影响

     

摘要

Aim:To clear whether Neonatal maternal separation (NMS)changes adulthood glucose metabolism and increases the risk of type 2 diabetes caused by high fat diet.Methods:A rat NMS model was used and adult rats were divided into 4 groups.After adult rats were feed by high fat diet for 2 months,the body weights,oral glucose tolerance and blood pressures were detected.Results:The weights of NMS adult rats and NH rats have no significant difference,but the weights of NMS rats were higher than NH rats with the growth of the rat.Adult rats feed with high fat diet for a month or two months,the weights of NH rats and NMS rats with high-fat diet were higher than NH rats and NMS with normal diet,respectively.Fasting blood glucose values of adult NMS rats were decreased compared to a-dult NH rats while the blood glucose values of NMS rats were higher than that of NH rats after 30 min ad-ministered with glucose orally.The blood glucose values of NH rats with high fat diets,NMS rats with normal diets or high fat diets were higher than that of NH rats with normal diets after 60 min,90 min and 120 min administered with glucose orally.The blood glucose values of NMS rats with high fat diets were higher than that of NMS rats with high fat diets after 90 min and 120 min administered with glucose oral-ly.The blood pressure values had no significant difference between groups.Conclusion:NMS may induce the impaired glucose tolerance and the disorders of glucose metabolisms of rats.NMS may increase the onset sensitivities of type 2 diabetes.%目的:明确新生期母婴分离是否改变成年期糖代谢及增加高脂饮食导致2型糖尿病的风险.方法:建立大鼠NMS模型,正常(NH)大鼠和NMS大鼠成年后高脂饮食干预2个月,检测大鼠体重、口服葡萄糖耐量和血压.结果:NMS大鼠成年后体重与NH大鼠体重无明显差别,但随着鼠龄的增长,NMS大鼠体重较NH大鼠体重增加;成年后大鼠经高脂饮食干预1个月或2个月,高脂饮食组NH和NMS大鼠分别较正常饮食组NH大鼠和NMS组体重明显增加;成年后NMS大鼠空腹血糖较NH大鼠低,但NMS大鼠口服葡萄糖后30 min的血糖值较NH大鼠高;NMS正常饮食组、NH高脂饮食组和NMS高脂饮食组口服葡萄糖后60、90、120 min的血糖值均比NH正常饮食组高;NMS高脂饮食组口服葡萄糖后90、120 min的血糖值均比NMS正常饮食组高;各组大鼠血压均无明显差异.结论:NMS可能导致大鼠糖耐量减低,体内糖代谢紊乱,可能增加发生2型糖尿病的敏感性.
