凋亡调控蛋白bcl-2、caspase-3在大鼠多囊卵巢颗粒细胞中的表达及生物学意义

         

摘要

Objective:To explore the expression of bcl-2 and caspase-3 apoptosis protein in rat's different levels of follicle ovary granular cells with polycystic ovary syndrome. To study granular cell's effect in PCOS's occurrence and development,in order to offer the theory basis of mechanism in PCOS. Methods:( 1 ) Establishing animal model of PCOS by the combination method of progestogen with chorionic gonadotropin hormone(HCG). (2)Using immunohistochemistry to explore the expression of bcl-2 and caspase-3 protein betwe en the PCOS group and normal group in different levels of follicle cells. Results:The bcl-2 protein expresses between the model group and normal group in different levels follicle cytoplasm. There is a distinguish difference among the primordial and primary and secondary follicle in model group(P<0. 05). The caspase-3 protein expresses between the model group and normal group in different levels follicle nucleus and cytoplasm. There is a distinguish difference among the primordial and primary and secondary follicle in normal group ( P<0 . 05 ) . There is a distinguish difference among primordial and primary and mature follicle in model group of secondary follicle( P<0. 05 ). Comparing the model group with normal group in follicle granular cells, the expression of bcl-2 protein was decrease and caspase-3 protein were higher. Conclusion:The pathogenesis of POCS may have related with the excessive apoptosis of granular cells.%目的::研究凋亡调控蛋白bcl-2、caspase-3在多囊卵巢综合征( polycystic ovary syndrome,PCOS)大鼠卵巢颗粒细胞中的表达情况,探讨颗粒细胞凋亡在PCOS发生、发展中的作用,为进一步阐述PCOS的发病机制提供理论依据。方法:(1)采用孕激素联合HCG法建立多囊卵巢综合征大鼠模型。(2)采用免疫组化SP法检测bcl-2、caspase-3在PCOS组和正常组各级卵泡中的表达。结果:bcl-2在模型组和正常组卵巢的各级卵泡的颗粒细胞的胞浆表达。模型组原始卵泡、初级卵泡、次级卵泡与成熟卵泡有显著性差异(P<0.05);caspase-3在模型组和正常组卵巢的各级卵泡的颗粒细胞的胞核和胞浆中均有表达。正常组中原始卵泡与初级卵泡、次级卵泡、成熟卵泡有显著性差异(P<0.05);模型组中次级卵泡与原始卵泡、初级卵泡、成熟卵泡有显著性差异(P<0.05)。模型组与正常组比较卵泡颗粒细胞中 bcl-2蛋白表达降低,模型组与正常组比较卵泡颗粒细胞中caspase-3蛋白的表达升高。结论:多囊卵巢综合征与颗粒细胞的过度凋亡有关。
