两种比色法测定荞麦降糖颗粒中总黄酮的含量

         

摘要

目的::通过比较 NaNO2-Al( NO3)3-NaOH 法和 AlCl3法的差异,建立荞麦降糖颗粒中总黄酮含量的比色分析方法。方法:以乙醇为溶剂进行回流提取,并以芦丁为对照品,分别以 NaNO2-Al ( NO3)3-NaOH 和AlCl3为显色剂,分别在516nm和417nm处用可见分光光度法测定荞麦降糖颗粒中总黄酮含量。结果:建立硝酸铝法芦丁浓度-吸光度关系的回归方程方程式 y=0.01201x-0.0291,R2=0.9994,回收率为100.2%。同时也建立三氯化铝法芦丁浓度-吸光度关系的回归方程方程 y=0.02992+0.00473,R2=0.9996,回收率为99.75%。结论:AlCl3法操作简便快速、专属性强,更适用于荞麦降糖颗粒中总黄酮的含量测定。%Objective:By comparing the difference between the NaNO2 -Al ( NO3 ) 3 -NaOH methods and AlCl3 methods,to set up a Colorimetric assay for the determination of total flavones in the Buckwheat Hypoglycemic granule. Methods:Extract the total flavones by ethanol, with rutin as standard sample,NaNO2-Al( NO3 ) 3-NaOH and AlCl3 as color reagents,the flavonoids content was de-termined by VlS-spectrophotometry at the wavelength of 516nm and 417nm. Result:The regression equation of NaNO2-Al(NO3)3-NaOH methods was y=0. 01201x-0. 0291,R2=0. 9994,the average recovery of this method was 100. 2%. The regression equation of AlCl3 methods was y=0. 02992 x +0. 00473,R2=0. 9996,the average recovery of this method was 99. 75%. Conclusion:The method of spectmphotometry in the AlCl3 is simple, rapid, highly accurate, and suitable for quality control of Buckwheat Hypoglycemic granule.
