前癃通胶囊抑制良性前列腺增生基质细胞TGF-β1表达的体外实验研究

     

摘要

目的:研究前癃通胶囊干预下,TGF-β1mRNA在良性前列腺增生基质细胞中的表达的变化.方法:采用MTT比色法检测前癃通胶囊对体外培养良性前列腺增生前列腺基质细胞增殖的抑制作用;应用逆转录-多酶链反应(RT-PCR)技术,观察前癃通胶囊干预下TGF-β1mRNA在良性前列腺增生基质细胞中的表达的变化.结果:3种浓度的前癃通胶囊能抑制前列腺基质细胞的增殖,且随着时间的延长抑制作用增强,高浓度组在不同时段对前列腺基质细胞增殖有明显的抑制作用,与中浓度组比,组间比较差异有统计学意义(P<0.01);中浓度组在不同时段对前列腺基质细胞增殖有抑制作用,与低浓度组比,组间比较差异有统计学意义(P<0.01).前列腺基质细胞中的TGF-β1mRNA表达随着前癃通胶囊(药液)浓度的增高呈现下降趋势.TGF-β1 mRNA在高浓度组的表达最低,空白组的表达最高.高、中、低浓度组分别与空白组比较差异有统计学意义(P<0.01);高、中浓度组与低浓度组比较TGF-β1mRNA的表达明显减弱,差异有统计学意义(P<0.01).结论:前癃通胶囊对前列腺基质细胞的增殖有较强的抑制作用,能降低TGF-β1 mRNA在基质细胞中的表达.%Objective To investigate the influence of Qianlongtong capsules on TGF-β1 Mrna expression in benign prostatic hyperplasia (BPH) prostatic matrix cells from human cultured in vitro. Methods The influence of Qianlongtong capsules in different concentrations on the proliferation of BPH prostatic matrix cells was tested by MTT assay. The influence of Qianlongtong capsules in different concentrations on the expression of TGF-β1 Mrna in BPH prostatic matrix cells was observed by RT-PCR. Results The proliferations of BPH prostatic matrix cells were inhibited by Qianlongtong capsules in these three concentrations,and the inhibition effects of Qianlongtong capsules enhanced with time. Compared with the medium-concentration group, there were obvious inhibition effects in Qianlongtong capsules with high concentration group at different phase and there were statistically significances (P<0.01). And compared with the low-concentration group, there were significantly inhibition effects in Qianlongtong capsules with medium concentration group (P< 0.01). The expression levels of TGF-B1 Mrna in cultured cells were decreased as increasing concentration of the Qianlongtong capsules. TGF-pi Mrna expression levels were lowest in high -concentration Qianlongtong capsules and highest in blank group. Compared with the blank group, TGF -β1 Mrna expression levels were statistically decreased in Qianlongtong capsules high-concentration group, medium-concentration group and low-concentration group (P< 0.01). Compared with the low-concentration group, TGF-fJl Mrna expression levels were statisti- cally decreased in Qianlongtong capsules high-concentration group and medium-concentration group (P<0.01). Conclusion Qianlongtong capsules have strong inhibition effects on the proliferation of BPH prostatic matrix cells, and can reduce the expression of TGF-β1 Mrna in the cells.
