自发性高血压大鼠左心室肥厚的代谢组学研究

         

摘要

目的 以血压正常的wistar大鼠为对照,研究自发性高血压大鼠(spontaneously hypertensive rats,SHR)左心室肥厚心肌组织的小分子产物变化.方法 采用气相色谱-质谱联用仪(GC-MS),对组织代谢产物进行检测分析;应用偏最小二乘法判别分析(partial least squares discriminant analysis,PLS-DA)分析差异代谢产物谱.依据变量重要性(VIP)、自变量系数探索SHR发生左心室肥厚心肌时代谢产物.结果 与SHR组呈正相关的是:90角鲨烯、51对苯二甲酸、87麦芽糖、68棕榈油酸、77葡萄糖-6-磷酸1、82-羟基丁酸、78未知、74油酸、56果糖1、28三氰丙氨酸;负相关的是:21琥珀酸、24富马酸、2丙酮酸、34天门冬氨酸1、85未知、45未知、88甘油单油酸酯、86油精.结论 进一步证实高血压导致的左心室肥厚与心肌细胞结构破坏以及能量失衡密切相关.%Objective To investigate the changes of small molecular products in left ventricular hypertrophyof spontaneously hypertensive rats (SHR), the Wistar rats with normal pressure as the control group. Methods The metabolites were detected by gas chromatography-mass spectrometry (GC-MS), and the differences of metabolite were analyzed by partial least squares (partial least squares discriminant analysis, PLS-DA). According to the variable importance (VIP) and independent variable coefficients, the left ventricular hypertrophy and myocardial age products in SHR rats were investigated. Results SHR group was positively correlated with: 90 squalene, 51 terephthalic acid, 87 maltose, 68 palmitoleic acid, 77 glucose-6-phosphate 1, 8 2-hydroxybutyric acid, 78 unknown, 74 oleic acid, 56 fructose 1, 28 three cyanogens alanine. Negative related with: 21 succinate, 24 fumarate, 2 pyruvic acid, 34 aspartic acid 1, 85 unknown, 45unknown, 88 glycerol ester of single oil, 86 olein. Conclusion It is further confirmed that the left ventricular hypertrophy induced by hypertension is closely related to the destruction of myocardial cell structure and the imbalance of energy.
