丁苯酞-羟丙基-β-环糊精包合物的制备与表征

             

摘要

The butylphthalide-hydroxypropyl-β-cyclodextrin inclusion compound was prepared by freeze-drying method, and the encapsulation efficiency was determined. The inclusion complex was characterized by thin layer chromatogram (TLC), ultraviolet spectroscopy( UV) , infrared spectroscopy(IR), differential scanning calorimetry(DSC) and thermal gravimetric analy-sis(TGA). The results show that the TLC,UV,IR,DSC and TGA method can be used to characterize the formation of the inclusion compound. The inclusion compound is generated under 70 ℃. The reaction time is 4 hours, and the mole ratio of hydroxypropyI-β-cyclodextrin to butylphthalide is 1 ∶ 1. Inclusion entrapment efficiency is up to 65. 22%. The complexation constant calculated from phase diagram is 268. 99, and the inclusion compound could increase solubility by 108. 4 times.%设计正交试验确定冷冻干燥法制备丁苯酞—羟丙基-β-环糊精包合物的最佳条件,通过薄层色谱法(TLC)、紫外光谱法(UV)、红外光谱法(IR)、差示扫描量热法(DSC)、热重分析法(TGA)考察所制备的丁苯酞与羟丙基-β-环糊精(HP-β-CD)包合前后的变化.结果表明:TLC,UV,IR,DSC,TGA可以表征包合物的形成,丁苯酞与HP-β-CD在70℃、反应4h所形成的包合物主、客体分子物质的量比为1∶1,包封率为65.22%,包合物的表观稳定常数为268.99.对包合前后的水溶解性考察结果表明,形成包合物后可以使丁苯酞增溶108.4倍.
