KCTD-12抑制胃间质瘤细胞体外增殖、侵袭的实验研究

         

摘要

Objective: To study the effects of potassium channel tetramerization domain-containing 12 (KCTD-12) overexpression on in vitro proliferation and invasion of gastrointestinal stromal tumor cells.Methods: Gastrointestinal stromal tumor cell lines were cultured and randomly divided into KCTD-12 overexpression group and NC overexpression group that were transfected with KCTD-12 overexpression plasmid and negative control plasmid respectively.24 hours after transfection, the expression of proliferation genes, apoptosis genes, PI3K and p38MAPK signaling pathway molecules as well as invasion genes in cells were detected.Results: Ki-67, SCIN, Bcl-2, p-PI3K, p-AKT, p-p38MAPK, CD146, PAR-2, SDF-1 and CXCR4 expression in KCTD-12 overexpression group were significantly lower than those in NC overexpression group while Bax, Caspase-3, Caspase-7, Caspase-9 and p-FOXO1 expression were significantly higher those in NC overexpression group.Conclusions: KCTD-12 can inhibit the in vitro proliferation and invasion of gastrointestinal stromal tumor cells.%目的:研究过表达钾通道四聚化结构域-12蛋白(KCTD-12)对胃间质瘤细胞体外增殖、侵袭的影响.方法:培养胃间质瘤细胞株GIST-T1并随机分为KCTD-12过表达组和NC过表达组,分别转染KCTD-12过表达质粒及阴性对照质粒.转染后24小时,检测细胞中增殖基因、凋亡基因、PI3K及p38MAPK信号通路分子、侵袭基因的表达量.结果:KCTD-12过表达组细胞中Ki-67、SCIN、Bcl-2、p-PI3K、p-AKT、p-p38MAPK、CD146、PAR-2、SDF-1、CXCR4的表达量显著低于NC过表达组,Bax、Caspase-3、Caspase-7、Caspase-9、p-FOXO1的表达量显著高于NC过表达组.结论:KCTD-12能够抑制胃间质瘤细胞的体外增殖、侵袭.
