骨肉瘤中FoxM1、Sox9基因异常表达与肿瘤病理特征的相关性研究

         

摘要

Objective:To study the correlation of the abnormal expression of FoxM 1 and Sox9 in osteosarcoma with the pathologic features of tumor.Methods:Patients with osteosarcoma and those with giant cell tumor who received surgical resec-tion in Enshi Youfu hospital between March 2015 and August 2017 were selected and enrolled in the osteosarcoma group and control group of the study after the tissue properties were confirmed by pathological examination.The lesion tissue was collect-ed after operation to determine the mRNA expression of FoxM 1,Sox9,proliferation genes and invasion genes.Results:FoxM1,Sox9,AURKA,PAK5,IRX2,S100A6,FAK,MMP9,PIPK4 and N-cadherin mRNA expression in lesion tissue of osteosarcoma group were significantly higher than those of control group whereas Id1 and TAp73 mRNA expression were sig-nificantly lower than those of control group;AURKA,PAK5 and IRX2 mRNA expression in lesion tissue of osteosarcoma group with high FoxM1 expression were significantly higher than those in lesion tissue with low FoxM 1 expression whereas Id1 and TAp73 mRNA expression were significantly lower than those in lesion tissue with low FoxM 1 expression;S100A6,FAK, MMP9,PIPK4 and N-cadherin mRNA expression in lesion tissue of osteosarcoma group with high Sox9 expression was signifi-cantly higher than those in lesion tissue with low Sox9 expression.Conclusion:The abnormal high expression of FoxM1 and Sox9 in osteosarcoma can promote the cell proliferation and invasion within the tumor.%目的:研究骨肉瘤中FoxM1、Sox9基因异常表达与肿瘤病理特征的相关性.方法:选择在恩施州优抚医院接受手术切除的骨肉瘤患者和骨巨细胞瘤患者,经病理检查明确组织性质后纳入研究的骨肉瘤组及对照组.手术后收集病灶组织并测定FoxM1、Sox9基因及增殖基因、侵袭基因的mRNA 表达量.结果:骨肉瘤患者病灶组织中 FoxM1、Sox9、AURKA、PAK5、IRX2、S100A6、FAK、MMP9、PIPK4、N-cadherin的mRNA表达量显著高于对照组,Id1、TAp73的mRNA 表达量显著低于对照组;骨肉瘤组中FoxM1高表达病灶组织中AURKA、PAK5、IRX2的mRNA表达量显著高于FoxM1低表达的病灶组织,Id1、TAp73的mRNA表达量显著低于FoxM1低表达的病灶组织;骨肉瘤组中Sox9高表达病灶组织中S100A6、FAK、MMP9、PIPK4、N-cadherin的mRNA 表达量显著高于Sox9低表达的病灶组织.结论:骨肉瘤中FoxM1、Sox9基因的异常高表达分别能够促进肿瘤病灶内细胞的增殖和侵袭.
