一种改进的贵阳腐霉保种方法

         

摘要

Objective:To improve the storage method for strain of Pythim guiyangense Su.Methods:The mycelia grew on SFE and water agar solid media,and agar blocks with mycelia were stored in aseptic distilled water at room temperature (SFE in 45 tubes,water agar in 183 tubes).The agar blocks were taken out from different tubes and planted on KPYG2 agar plates kept in incubator for mycelial growth on the 72nd,200th,250th,350th,400th,450th,930th,and 1008th days of the experiment respectively.The storage results in terms of living duration and mycelial status were compared.Results:Mycelia from 8 tubes of the 45 SFE tubes did not grow on the 72nd day,those from 4 tubes of SFE tubes kept alive on the 450th day,and those from other SFE tubes only be observed alive on the 200th day.In regard of the mycelia grown on water agar,the cultures from the 183 tubes were all alive in the subsequent tests.Although the tubes were reduced,there still were mycelia from 3 tubes alive on the 1008th day.The best results were obtained when the fungus grew on water agar plates and then immersed in disinfected double-distilled water,by which,generally the fungus kept alive for at least one year or above,and the longest storage duration of 1008-days was obtained.Conclusions:So far,distilled water immersing mycelia grown on water agar would be an ideal method for storage of P.guiyangense strains.%目的:改进贵阳腐霉菌种的保存方法.方法:采用葵花籽浸膏(SFE)和水琼脂固体培养基分别接种贵阳腐霉(SFE培养基45支,水琼脂固体培养基183支),切取生长有菌丝的琼脂块置无菌蒸馏水室温保存,于保存后第72、200、250、350、400、450、930及1 008天取出试管内琼脂块接种于KPYG2琼脂培养基,置恒温箱培养,观察菌丝生长情况,记录各试管中菌丝的存活时间.结果:盛放SFE琼脂块的45支试管,有8支于第72天检测未见菌丝生长,4支试管在第450天时检出存活菌丝,其余试管于第200天检出存活菌丝;培养到第72、200、350、400、450、930及1 008天时水琼脂固体培养基中还能检出存活的贵阳腐霉菌丝,试管里的水保持清澈透明,但管数随培养时间的延长有所下降,到第1008天检测仍然有3支菌种检出存活的贵阳腐霉菌丝.结论:水琼脂培养的贵阳腐霉置于无菌水中是目前比较好的保种方法.
