青蒿琥酯对人急性白血病原代细胞线粒体膜电位的影响

         

摘要

Objective To observe the effect of artesunate on mitochondrial membrane potential of human acute leukemia primary cells. Methods Bone marrow cells isolated from acute leukemia patients were cultured with artesunate at the final concentration of 50, 25, 12. 5 μg/mL in 6-well plate respectively. After culturing for 48 hours, the cultured cells were labeled with fluorescence probe JC-1 and then were fixed. We observed cell morphology under the laser scanning confocal microscope, and counted the cells with red fluorescence and the cells with green fluorescence. Results After treatment with artesunate at different concentrations, the number of cells with green fluorescence increased ( P < 0. 05 compared with that in the blank control group), and the number of cells with red fluorescence decreased to various extent ( P > O. 05 ). Conclusion The mechanism of artesunate for treatment of acute leukemia is related with the decrease of mitochondrial membrane potential of human acute leukemia primary cells, which results in the tumor cell death through inducing the changes of cellular internal environment.%[目的]观察青蒿琥酯作用于人急性白血病原代细胞后线粒体膜电位的变化.[方法]选取急性白血病患者的骨髓进行原代细胞培养,将终浓度为50、25、12.5μg/mL的青蒿琥酯分别作用于细胞,经线粒体膜电位荧光探针Jc-1染色并固定后,在激光扫描共聚焦显微镜下观察细胞形态的变化,并计算红、绿荧光细胞数.[结果]不同浓度青蒿琥酯作用于人急性白血病原代细胞后,均可升高绿荧光细胞数,与空白对照组比较差异有显著性意义(P<0.05),并可不同程度降低红荧光细胞数(P>0.05),且呈一定的剂量依赖关系.[结论]青蒿琥酯抗急性白血病原代细胞的作用机制与降低其线粒体膜电位,进而使其胞内环境改变而使肿瘤细胞凋亡有关.
