瑞香素对人子宫内膜癌细胞生物学功能的影响

     

摘要

目的:探讨瑞香素对人子宫内膜癌HEC-1A和HEC-1B细胞生物学功能的影响.方法:取对数生长期的人子宫内膜癌HEC-1A和HEC-1B细胞,分为对照组(0μmol/L瑞香素)和不同浓度(50μmol/L、75μmol/L、100μmol/L)瑞香素处理组,培养48h后,采用CCK-8法检测细胞增殖抑制率,流式细胞术AnnexinV-APC/PI双染法检测细胞凋亡率,划痕实验和Transwell细胞侵袭实验分别检测瑞香素对细胞体外迁移能力和侵袭能力的影响.结果:在HEC-1A和HEC-1B细胞中,不同浓度瑞香素处理组细胞增殖抑制率和迁移率均较对照组显著降低,Transwell细胞侵袭实验中的跨膜细胞数显著减少,而细胞凋亡率明显升高,差异均有统计学意义(均P<0.05),且不同浓度瑞香素处理组组间两两比较,差异亦有统计学意义(P<0.05).结论:瑞香素可抑制人子宫内膜癌HEC-1A和HEC-1B细胞增殖、侵袭和迁移,并诱导细胞凋亡,且呈剂量依赖关系.%Objective:To investigate the effects of daphnetin on the biological function of human endometrial carcinoma HEC-1Aand HEC-1Bcells.Methods:HEC-1Aand HEC-1 Bcells were divided into different concentrations(50μmol/L,75μmol/L and 100μmol/L) of daphnetin treatment groups and control group.The cell counting kit-8(CCK-8) method,AnnexinV-APC/PI double staining,scratch test and Transwell assay were used to detect the proliferation,apoptotic rate,migration and invasion ability of HEC-1A and HEC-1B cells,respectively.Results:The proliferation rate,migration rate and the number of transmembrane cells in Transwell assay were significantly decreased,while the apoptotic rate was notably increased in different concentrations of daphnetin groups,compared with the control group (P<0.05).There were significant differences among different concentrations of daphnetin treatment groups as to these indexes(P<0.05). Conclusion:Daphnetin could inhibit the proliferation,invasion and migration,and promote apoptosis of human endometrial carcinoma HEC-1A and HEC-1B cells,and the effect was dose-dependent.
