低剂量阿霉素富集肾上腺皮质癌干细胞的实验研究

摘要

Objective: To investigate the effective enrichment modles of adrenocortical cancer stem cells and its possible storting marker. Methods: Establish the enrichment modles, that SW-13 cells were continuously passaged in nude mice with adriamycin. Then the detection indicators of enrichment efficiency included Sphere-forming assay and Clony-forming assay. The expression of cancer stem cell related markers: CD184 CD338 was detected with flow cytometry. Results: The tumortake rate of all the nude mice was 100%. Statistically significance was different among the Sphere formation rate and the clone formation rate of cell blank control group, SW-13/ADM P1 , SW-13/ADM P2 ( P <0. 05) : SW-13/ADM P2 > SW-13/ADM P1 > blank control group. Compared with blank control group and SW-13/ADM P1 , the expression of CD184 CD338 of cell SW-13/ADM P2 was higher ( P <0. 05). Conclusion: In vivo, this enrichment models with adriamycin can effectively enrich the adrenocortical cancer stem cells. Cancer stem cell related markers: CD184 CD338 may be the possible storting marker.%目的:探讨肾上腺皮质癌干细胞的有效富集模式和其可能的表面标志物.方法:构建裸鼠皮下移植瘤反复传代联合化疗药物阿霉素干预的动物模型,富集人肾上腺皮质癌细胞系SW-13的干细胞,并进行富集效率检测("悬浮球"形成试验;平板克隆形成实验).流式细胞术检测肿瘤标志物组合CD184+CD338+,研究SW-13干细胞可能的表面标志物.结果:裸鼠皮下移植瘤成瘤率为100%.空白对照组及各代SW-13裸鼠皮下移植瘤细胞的"悬浮球"形成率、平板克隆形成率比较差异均有统计学意义(P<0.05),富集第2代细胞>第1代细胞>空白对照组.流式细胞术检测富集第2代细胞的CD184+CD338+表达率较第1代细胞与空白对照组增高(P<0.05).结论:裸鼠皮下移植瘤反复传代联合化疗药物阿霉素干预的富集模式,可有效富集肾上腺皮质癌SW-13干细胞样细胞.CD184+CD338+可作为肾上腺皮质癌SW-13干细胞的候选标志物组合.
