Objective:To investigate the effect of matrix metalloproteinases-10 (MMP-10) gene silencing on subcutaneous tumor of human lung adenocarcinoma cells in nude mice.Methods:Adopting lentivirus of siRNA which carry interference sequence to silence the expression of MMP-10 gene in A549 cells,building MMP-10 silence stable cells strain and nude-mouse transplanted tumor model,and comparing the differences of growth speed,size,and weight of nude mouse with subcutaneous tumor between the experimental group (A549-si MMP-10) and the control group (A549-NC).Results:The growth speed of tumor from A549-si MMP-10 group was significantly slower than the A549-NC group,as well as the size and weight (P<0.05).Conclusion:MMP-10 genes might relate to the occurrence and development of adenocarcinoma cells of human lung.%目的:探讨沉默基质金属蛋白酶-10(MMP-10)对人肺腺癌裸鼠皮下移植瘤的影响.方法:采用携带干扰序列的siR-NA慢病毒沉默A549细胞MMP-10的表达,构建沉默MMP-10表达的稳转细胞株及裸鼠移植瘤模型,比较实验组(A549-siMMP-10组)和对照组(A549-NC组)裸鼠成瘤瘤体生长速度、瘤体体积及瘤重的差异.结果:A549-si MMP-10组裸鼠成瘤瘤体生长速度明显慢于A549-NC组,瘤体体积及瘤重明显小于A549-NC组,差异均有统计学意义(均P<0.05).结论:MMP-10基因可能与人肺腺癌的发生、发展密切相关,MMP-10基因有望成为判断人肺腺癌预后的一个新的重要指标.
