白藜芦醇抗喘活性及机制研究

     

摘要

目的 观察白藜芦醇对豚鼠在体和离体气管平滑肌的松弛作用,并探讨其作用机制.方法 建立豚鼠哮喘模型,采用豚鼠离体气管条法,观察白藜芦醇对豚鼠气管平滑肌的舒张作用.结果 白藜芦醇可显著延长豚鼠引喘潜伏期,对乙酰胆碱、氯化钾、组胺诱发的气管收缩有明显的舒张作用;吲哚美辛和格列本脲预孵育明显减少了白藜芦醇对离体气管条的舒张作用.结论 白藜芦醇具有平喘和明显的舒张气管平滑肌的作用,其机制可能与钙拮抗、促进前列腺素合成和激活ATP敏感性钾离子通道有关.%Objective To study the relaxant effect of resveratrol ( Res) in vivo and in vitro on guinea-pig tracheal smooth muscle, and its potential mechanism. Methods Tracheal strips were isolated and the model of asthmatic guinea-pig was established. The function of Res on antiasthmatic effect was detected. Results Res obviously prolonged theifttency time of asphyxiation collapse induced by histamine (His) and acetyl-choline (Ach) in guinea-pigs, and abrogated the tension of tracheal smooth muscle of precontraction induced by Ach, KC1 and His. The effect of Res on relaxation was attenuated by pretreatment with indomethacin and glibenclamide. Conclusion Res exhibit a significant antiasthmatic and relaxing effect on tracheal smooth muscle of guinea-pig. The mechanism of Res may be related to antagonizing of calcium channel, elevating prostaglandin levels, and opening ATP-sensitive potassium channel.
