以红核子龙眼不同保存时间(5个月、3 a、13 a)的不同发育阶段体胚为材料,对限制生长保存后的胚性愈伤组织(EC)的体胚发生过程进行随机扩增多态性DNA (RAPD)分析;筛选10条随机引物,对EC不同发育阶段体胚的基因组DNA进行扩增.结果表明:龙眼EC不同发育阶段体胚的RAPD谱带存在一定的差异,但不同保存时间的EC在体胚发生过程中的相对变异率都保持在比较小的变异范围内,小于5.1%.%The hereditary stability was detected in different developmental-stage cultures by randomly amplified polymorphic DNAs (RAPD) technique (amplification with selected 10 primers) with the materials of embryogenic cultures during somatic embryogene-sis under minimal growth conservation(5 months,3 years,13 years) in Dimocarpus longan cv. Honghezi. The results of RAPD analysis showed that there were some differences in the patterns of RAPD in the embryogenic cultures at different developmental satges during somatic embryogenesis, but the variation rates for different time of minimal growth conservation kept in lower levels (less than 5.1%) .
