液相色谱示差折光法测定酒中的总糖和还原糖

         

摘要

Objective To establish a method for the determination of the total sugar and reducing sugar in the wine by high performance liquid chromatogram-refractive index detection method (HPLC-RID).Methods Various alcoholic drinks were diluted of certain multiple according to their categories.After the filtration,4 kinds of sugar were determined by HPLC-RID directly.The total sugar content of the sample was determined to be the sum of fructose,glucose,sucrose and maltose,in which,the content of fructose,glucose and maltose were the amount of reducing sugars.Results The peaks of 4 kinds of sugar appeared within 15 min.The recoveries were ranged from 94.1% to 99.9% for the target compound residues with 3 spiked levels of 1,2,4 g/L.The relative standard deviations (RSD) were less than 2.34%.The limits of detection (LOD) were about 0.1 g/L and limits of quantitation (LOQ) were about 0.4 g/L.Conclusion The proposed method is simple,rapid,and accurate,which is suitable for analysis of total sugar and reducing sugar in most wines.%目的 建立高效液相色谱-示差折光检测法(high performance liquid chromatogram-refractive index detection method,HPLC-RID)测定酒的总糖和还原糖的分析方法.方法 根据不同酒的类别和含糖量进行其相应倍数的稀释,过滤后利用HPLC-RID法测定酒样中的4种糖,其总和为其总糖量,而果糖、葡萄糖和麦芽糖的含量之和,为该酒样还原糖量.结果 本方法在15 min内完成4种目标化合物的分离分析.酒样的4种糖在1、2、4 g/L添加水平的回收率为94.1%~99.9%,相对标准偏差最高为2.34%,方法检出限约为0.1 g/L,定量限约为0.4 g/L.结论 该方法快速、简便、准确,适用于大多数酒类样品的总糖与还原糖测定.
