本研究探讨丙戊酸钠(valproic acid sodium,VPA)对Jurkat细胞增殖的抑制作用及对凋亡的影响.选择不同浓度的VPA处理Jurkat细胞,采用CCK-8法检测并绘制增殖抑制曲线,Annexin V/PI双染流式细胞术检测细胞凋亡,半定量RT-PCR检测抑凋亡基因BCL-2及促凋亡基因Bak1的变化.结果表明,VPA对Jurkat细胞增殖的抑制作用呈浓度依赖性,与对照组相比,随药物浓度增高,细胞凋亡率增加；VPA能够使抑凋亡基因BCL-2表达减低,但对促凋亡基因Bak1的表达无明显影响.结论:VPA可促进Jurkat细胞凋亡,可能与其抑制BCL-2表达有关.%This study was aimed to investigate the effects of valproic acid sodium (VPA) on the proliferation and apoptosis of acute T-lymphoblastic leukemia Jurkat cells. Jurkat cells were treated with different concentration of VPA. Proliferation-inhibition curve was assayed and plotted by CCK-8 method and the cell apoptosis was detected by flow cy-tometry with Annexin V/PI double staining. The expression level of anti-apoptotic gene BCL-2 and pro-apoptosis gene Bakl were detected by semi-quantitive RT-PCR. The results showed that the VPA inhibited the proliferation of Jurkat cells in concentration-dependent manner. As compared with the control group, the apoptosis of cells increased along with adding concentration of VPA; VPA could decrease the expression of BCL-2 gene, but did not show obvious effect on the expression of Bakl. It is concluded that the VPA can inhibit proliferation of Jurkat cells which possibly associates with the decrease of BCL-2 expression.
