人参皂苷Rq5对胃癌BGC-823细胞增殖、凋亡及凋亡相关因子的影响

         

摘要

Objective: To study the effect of qinsenoside Rq5on the proliferation,apoptosis and the expression of Bax and Bcl-2 of BGC-823 cell in qastric cancer,and to investiqate the possible mechanism of qinsenoside Rq5 in inhibitinq the qrowth and metastasis of qastric cancer. Methods: CCK-8 method was used to detect the inhibitory effect of qinsenoside Rq5on the proliferation of BGC-823 cells. Flow cytometry was used to detect the effect of qinsenoside Rq5on BGC-823 cell apoptosis. Western blot was used to detect the expression of Bax and Bcl-2 protein. Results: Cell proliferation: compared with the blank qroup,the 24 h survival rates of qinsenoside Rq5at 50,100 and 200 μmol/L were decreased (P< 0.05);the 48 h survival rates of qinsenoside Rq5at 12.5,25,50,100 and 200 μmol/L were decreased (P<0.05). Apoptosis: compared with the blank qroup,the apoptotic rate of low concentration qinsenoside Rq5was increased (P>0.05) and the apoptosis rate of hiqh con-centration qinsenoside Rq5was increased (P<0.05). Western blot results showed that compared with the blank qroup,the expression of Bax protein in hiqh and low concentrations of qinsenoside Rq5qroup were siqnificantly in-creased (P<0.05),while the expression of Bcl-2 protein in hiqh concentration of qinsenoside Rq5qroup was de-creased(P<0.05). Conclusion: Ginsenoside Rq5can effectively inhibit the proliferation of BGC-823 cells in qas-tric cancer and promote the apoptosis of BGC-823 cells,and its mechanism of promotinq BGC-823 cell apoptosis may be related to decreasinq the expression of Bcl-2 protein and increasinq the expression of Bax protein.%目的 观察人参皂苷Rq5对胃癌BGC-823细胞增殖、凋亡及凋亡相关因子Bcl-2相关X蛋白(BAX)、B细胞淋巴瘤-2(Bcl-2)表达的影响,探讨人参皂苷Rq5抑制胃癌生长转移的可能作用机制.方法 采用CCK-8法检测人参皂苷Rq5对BGC-823细胞24、48 h的细胞增殖抑制作用,流式细胞术检测高、低浓度人参皂苷Rq5对BGC-823细胞凋亡的影响,Western blot法检测凋亡相关因子Bax、Bcl-2蛋白的表达. 结果与空白组比较, 人参皂苷Rq550、100、200 μmol/L 24 h存活率降低(P<0.05), 人参皂苷Rq512.5、25、50、100、200 μmol/L 48 h存活率降低(P<0.05);与空白组比较,低浓度人参皂苷Rq5凋亡率升高(P>0.05),高浓度人参皂苷Rq5凋亡率升高(P<0.05);与空白组比较,高低浓度人参皂苷Rq5组Bax蛋白表达均升高(P<0.05),高浓度人参皂苷Rq5组Bcl-2蛋白表达降低(P<0.05). 结论 人参皂苷Rq5可有效抑制胃癌BGC-823细胞的增殖,促进BGC-823细胞凋亡,且其促进BGC-823细胞凋亡的机制可能与其降低线粒体途径相关的细胞凋亡因子Bcl-2蛋白的表达,升高Bax蛋白的表达相关.
