钩芍降压颗粒对自发性高血压大鼠血管紧张素Ⅱ及肾动脉钙敏感受体的影响

摘要

目的 研究钩芍降压颗粒对自发性高血压大鼠血管紧张素Ⅱ及肾动脉钙敏感受体的影响,探讨钩芍降压颗粒可能的作用机制.方法 取雄性自发性高血压大鼠30只,随机分为模型对照组、阳性对照组及试验组,每组10只,另雄性Wistar大鼠10只作为空白对照组.阳性对照组予厄贝沙坦,试验组钩芍降压颗粒、模型对照组与空白对照组予等量蒸馏水.每日1次,连续4周.每周检测血压、酶联免疫吸附法测定血清血管紧张素Ⅱ含量、免疫组化法检测钙敏感受体表达.结果 与阳性对照药比较,钩芍降压颗粒起效慢、作用轻微.血清血管紧张素Ⅱ含量,模型对照组、阳性对照组与空白对照组比较,差异有显著统计学意义(P＜0.01);试验组与空白对照组、阳性对照组比较,差异无统计学意义(P＞0.05).肾动脉钙敏感受体表达,模型对照组与空白对照组比较差异无统计学意义(P＞0.05);阳性对照组空白对照组比较差异有统计学意义(P＜0.05);试验组与空白对照组比较差异有显著统计学意义(P＜0.01);试验组与阳性对照组比较差异无统计学意义(P＞0.05).结论 钩芍降压颗粒能降低自发性高血压大鼠血清血管紧张素Ⅱ含量,为其降血压作用机制之一.可能通过活化肾动脉钙敏感受体缓解“肾动脉痉挛”而降低AngⅡ含量,但需进一步研究证实.%Objective:To research the influence of Goushao Jiangya Granule on Angiotensin Ⅱ and renal artery calcium sensing receptor in spontaneously hypertensive rats.Methods:30 SHR were randomly divided into the model control,the positive control group and the experimental group,another 10 Wistar rats as the blank control group.The positive control group was gave Irbesartan Tablets.The experimental group was gave Goushao Jiangya Granule.The model control group and the blank control group were given equal distilled warter.It lasted for 4 weeks,once per day.Blood pressure was detected weekly.Enzyme-linked immunosorbent assay was used to detect serum angiotensin Ⅱ,and the expression of calcium sensing receptor was detected by immunohistochemistry.Results:Compared with the positive control drugs,Goushao Jiangya Granule had slow onset and a mild effect.There was a significant difference in serum angiotensin Ⅱ level between the model control group and the positive control group,compared with the blank control group (P＜0.01).There was no significant difference among the experimental group,the blank control group and the positive control group (P＞0.05).The expression of calcium sensitive receptor in renal artery was not significantly different between the model control group and the blank control group (P＞0.05).The difference between the positive control group and the blank control group was statistically significant (P＜0.05).The difference between the experimental group and the blank control group was statistically significant(P＜0.01),and the difference between the experimental group and the positive control group was not statistically significant(P＞0.05).Conclusion:Goushao Jiangya Granule can reduce serum angiotensin Ⅱ level in spontaneously hypertensive rats,and is one of its mechanisms of lowering blood pressure.It may reduce the level of Ang Ⅱ by activating the renal artery calcium sensing receptor to alleviate the "renal artery spasm",but we need further research to confirm that.
