Rad18缺失对雄小鼠生殖细胞的影响

         

摘要

[Objective] To investigate the function of Rad 18 in germ cells of male mice. [ Methods ] Use germ line cell -specific marker TRA98 to detect the germ cells in the seminiferous tubules of 2 - ,6 - and 12 - month - old Rad 18 -/ -mice. Use in situ hybridization to confirm the expression of Rad 18 in the spermatogonia of 1 - week wild type (WT) mice. [Results] The percentage of abnormal seminiferous tubules without spermatogonia in 12 -month - old Rad 18 -/ - testes was 39.5% ,more than 2.76% in WT testes, P <0.05. The degenerated tubules in Radl8 -/ - testes retained normal Sertoli cells. No TRA98 positive germ cells entered the Rad 18 -/- epididymis. The spermatozoa found in the 12 -month -old Radl8 -/ - epididymis displayed normal morphology. Radl8 was highly expressed in the spermatogonia of 1 -week -old WT mice. [Conclusion] Kad18 is involved in maintaining spermatogonial stem cells.%[目的]了解Rad18敲除雄小鼠生殖细胞的变化.[方法]用免疫组织化学方法检测2、6、12月龄Rad18-/-雄小鼠生精小管生殖细胞的特异性标记物TRA98的表达及分布,并与野生(WT)雄小鼠比较;用原位杂交技术检测Rad18在1周龄WT雄小鼠生精小管未分化精原细胞中的表达.[结果]缺失TRA98标记的早期精原细胞的生精小管比例在12月龄Rad18 -/-和WT雄小鼠中分别为39.50%和2.76%,差异有显著性意义(P<0.05);但Rad18 -/-退化生精小管中保留正常的Sertoli细胞,其附睾中未见TRA98阳性的生殖细胞,且精子形态正常;1周龄WT小鼠睾丸未分化精原细胞中有Rad18蛋白表达.[结论]Rad18可能对精原干细胞有维持作用.
