目的 探讨尿多酸肽(CDA-2)对慢性髓性白血病(CML) K562细胞增殖及凋亡的影响.方法 通过台盼蓝拒染法、吖啶橙(AO)荧光染色及流式细胞术,分别观察CDA-2处理前后K562细胞的增殖及凋亡情况.结果 台盼蓝拒染法显示,CDA-2可抑制K562细胞增殖,抑制作用在一定范围内呈剂量和时间依赖性,且CDA-2在31.25 mg/L以下浓度时,对K562细胞生长抑制作用较弱;AO荧光染色显示,经药物处理的K562细胞出现皱缩变小、核固缩呈均匀的致密物,且细胞碎片增多;流式细胞术显示,随着药物浓度的增加及作用时间的延长,存活细胞逐渐减少,凋亡细胞及死亡细胞逐渐增加.结论 CDA-2在体外可抑制K562细胞的增殖,并促进其凋亡.%Objective To explore the effects of uroacitide (CDA-2) on proliferation and apoptosis of K562 cells in chronic myeloid leukemia (CML).Methods By trypan blue stain assay,acridine orange (AO) fluorescent staining and flow cytometry,the proliferation and apoptosis of K562 cells before and after being dealt with CDA-2 were observed respectively.Results Trypan blue stain assay showed that CDA-2 can inhibit K562 cell proliferation,its inhibitory effect was dose-and timedependant within a certain range,and it exerted a weaker inhibitory effect on the growth of K562 cells when the concentration was less than 31.25 mg/L.AO fluorescent staining revealed that K562 cells dealt with drugs shrank and lessened,karyopyknosis with symmetrical dense deposits,and cell debris increased.Flow cytometry showed that living cells gradually decreased,and apoptotic and dead cells gradually increased with increased drug concentration and prolonged action time,.Conclusion CDA-2 can inhibit the proliferation of K562 cells in vitro and can promote fasten apoptosis.
