CD4+CD25+调节性T细胞及TGF-β1与系统性红斑狼疮病情活动性关系的研究

         

摘要

目的 检测系统性红斑狼疮患者(SLE)外周血CD4+CD25+调节性T细胞、转化生长因子-β1(TGF-β1)浓度,探讨其与疾病活动性的关系.方法 选择SLE患者76例,根据患者病情程度分为3组,其中稳定组17例,活动组33例,肾病组26例.同期选择24例健康体检者作为正常对照.采用流式细胞仪检测SLE患者外周血CD4+CD25+调节性T细胞群比率,用ELISA法检测血清中TGF-β1浓度.观察CD4+CD25+T细胞群、TGF-β1浓度与SLE患者疾病活动程度的关系.结果 ①活动期SLE患者外周血CD4+ CD25 +调节性T细胞群比率、TGF-β1浓度显著低于稳定期及正常对照组(P<0.01),疾病稳定期与正常对照组结果差异无统计学意义.②SLE并发肾病组外周血CD4+CD25+调节性T细胞群比例、TGF-β1浓度显著低于非肾病组(P<0.05).结论 SLE患者外周血CD4+CD25+调节性T细胞群比率、TGF-β1浓度与SLE活动性关系密切,可能是导致疾病发生和病情发展的关键环节之一.%Objective To study the percentage of peripheral blood CD4 + CD25 + regulative T cells and serum level of TGF - β1 in patients with systemic lupus erythematosus ( SLE ) and to explore their relationship with activity of the disease. Methods The percentage of CD4 + CD25 + T cells in peripheral blood from patients with systemic lupus erythematosus ( SLE ) and normal controls was measured by flowcytometry. Serum level of TGF - β1 was detected by double antibody sandwich ELISA. Results Patients with active disease had statisitically lower levels of CD4 + CD25 + T cells and TGF - β1 than those of normal controls and patients with non - active disease ( P < 0. 01 ), whereas no significant difference could be found between patients with non - active disease and normal controls. SLE patients with nephropathy had significantly lower levels of CD4 + CD25 + T cells and TGF - |31 than those of patients without nephropathy ( P <0101 ). Conclusion The decrease of percentage of peripheral blood CD4 + CD25 + T cells and serum level of TGF - (31 in patients with SLE is closely related with the activity of disease and complicated nephropathy. The decrease in percentage of blood CD4 + CD25 + T cells and serum level of TGF - (31 may play a crucial role in pathogenesis and progress of SLE.
