整合素 α7在肝癌组织中上调表达及对肝癌细胞增殖和迁移的影响

摘要

目的 检测整合素α7((ITGA7)在肝癌组织中的表达,研究消减整合素α7对肝癌细胞Hep3B增殖、迁移的影响.方法 检测ITGA7在56例肝癌患者癌组织和对应的癌旁组织中的表达情况,分析ITGA7的表达与肝癌临床、病理参数的关系.分别采用细胞克隆形成实验和细胞划痕损伤实验,检测消减ITGA7表达对Hep3B细胞的克隆形成、迁移能力的影响.结果 ITGA7在58.92％的肝癌组织中显著上调表达,肝癌组织中ITGA7的表达量与肝癌肿瘤大小(χ2=5.987,P=0.014))、有无包膜侵犯(χ2=9.402,P=0.0022))正相关.在Hep3B细胞中消减ITGA7的表达,能明显抑制细胞克隆形成和细胞迁移.结论 整合素 α7可能参与了肝癌的发生发展过程,在肝细胞恶性转化中发挥重要作用.%Objective To explore the expression of Integrin alpha 7 (ITGA7)in human hepatocellular carcinoma cells (HCC),and to study the effect of knocking down of ITGA7 on the proliferation and migration of HCC cell line. Methods The expression of ITGA7 was detected in tissues of HCC and corresponding neighboring normal tissues (56 samples),and its correlation with clinicopathological features had been ana-lyzed. Colony forming assay and scratch wound migration assay were used to explore the effect of knocking down of ITGA7 on proliferation and mi-gration of Hep3B cells. Results The expression of ITGA7 was increased in 58. 92％ (33 / 56)of HCC samples compared with corresponding neighboring normal tissues Moreover,the expression of ITGA7 had been found to be correlated with the size of tumor (χ2 = 5. 987,P = 0. 014) and the encapsulation of tumor (χ2 = 9. 402,P = 0. 0022). Knocking down of ITGA7 can inhibit the proliferation and migration of Hep3B cells. Conclusion Upregulation of Integrin α 7 can promote the proliferation and migration of hepatocellular carcinoma,and it is important in the process of pathogenesis of hepatic carcinoma.
