菩人丹超微粉对糖尿病大鼠视网膜TNF-α表达的影响

     

摘要

Objective To investigate the effects of Purendan superfine powder(PRD) on retinal tumor necrosis factor-a (TNF-α) expression in diabetic rat. Methods 36 male Wistar rats were randomly divided into 3 groups (n =12): normal control group, diabetes model group,PRD treatment group. The diabetes model rats were made by continuous intraperitoneal injection of Streptozotocin (STZ). After the diabetes animal model was successfully established, the rats in PRD treatment group were lavaged with PRD( 1.8 g·kg-1·d-1) for 3 months. The morphological changes of retina were observed by HE staining. Western blot were used to assess the expression of TNF-a protein in retina, reverse transcription-polvmerase chain reaction (RT-PCR) was employed to examine the expression of TNF-α mRNA in retina. Results In diabetes model rats,the retina showed obvious pathological changes,the expression of TNF-a protein and mRNA in retina were obviously higher than normal control rats (P < 0.01). Compared with diabetes model rats,the pathological changes of retina were alleviated significantly, the expression of TNF-α protein and mRNA in retina of rats in PRD treatment group decreased significantly (P < 0.01). Conclusion PRD can reduce vascular leakage and inhibit pathologic neovascularization by down-regulating TNF-α expression in retina,thus has protective effects on retinal microangiopathy during diabetes.%目的 探讨菩人丹超微粉( PRD)对糖尿病大鼠视网膜肿瘤坏死因子α(TNF-α)表达的影响.方法 36只Wistar大鼠随机分为正常对照组、糖尿病模型组和PRD治疗组,每组12只.糖尿病模型组、PRD治疗组大鼠均采用链脲佐菌素连续腹腔注射建立2型糖尿病大鼠模型.模型建立成功后,PRD治疗组大鼠给予PRD( 1.8 g·kg-1·d-1)灌胃3个月.HE染色观察视网膜的形态变化;采用Western blot法检测视网膜TNF-α蛋白的表达;逆转录聚合酶链反应检测视网膜TNF-α mRNA的表达.结果 糖尿病模型组大鼠视网膜出现明显的病理变化,TNF-α蛋白及mRNA表达显著高于正常对照组大鼠(P<0.01).PRD治疗组大鼠视网膜的病理变化明显减轻,TNF-α蛋白及mRNA表达显著低于模型组大鼠(P<0.01).结论 PRD可通过下调TNF-α表达,减少视网膜微血管渗漏及病理性新生血管生成,发挥对糖尿病视网膜微血管损伤的保护作用.
