喉鳞状细胞癌中STK15蛋白和BUBR1蛋白的表达及相互作用

         

摘要

Abstract Objective To explore the association of protein expressions of STK15 and BUBR1 with laryngeal squamous cell carcinoma (LSCC),and interaction between STIC 15 and BUBR1. Methods The laryngeal squamous cell carcinoma tissues and paired normal tissues were taken during operation from 40 patients without previous chemotherapy or radiotherapy. STK15 and BUBR1 protein expressions were evaluated by SP immunohistochemistry method. The associated protein expressions of STK15 and BUBR1 were evaluated by immunofluores-cence. Results The STK15 protein expression level of LSCC tissues was higher than that of paired normal tissues (P < 0.05),but the BUBR1 protein expression level was lower (P < 0.05). There might be an interaction between STK15 and BUBR1. Conclusion The results suggest that over-expression of STK15 protein and low-expression of BUBR1 protein might play a role in tumorigenesis and progression of Lscc. Besides,there might be an interaction between STK15 and BUBR1 ,but the specific mechanism needs further investigations.%目的 探讨STK15和BUBR1蛋白的表达与喉癌发生、发展的相关性以及喉癌组织中STK15和BUBR1蛋白的相互作用.方法选取40例病理诊断为喉鳞状细胞癌组织标本及癌旁正常组织标本,应用免疫组织化学SP法检测STK15和BUBR1蛋白的表达水平,应用免疫荧光双标实验判定STK15和BUBR1蛋白的共定位.结果STK15蛋白平均光密度值喉癌组织高于癌旁正常组织(P<0.05),BUBR1蛋白平均光密度值喉癌组织低于癌旁正常组织(P<0.05),即喉癌组织中STK15蛋白表达水平高于癌旁正常组织,而BUBR1蛋白表达水平低于癌旁正常组织.免疫荧光双标结果表明STK15蛋白和BUBR1蛋白均在细胞质及细胞核中表达,且2种蛋白标记的荧光可以重叠,提示两者可能存在相互作用.结论STK15蛋白高表达、BUBR1蛋白表达下调可能与喉癌的发生、发展相关.STK15和BUBR1蛋白可能相互作用,共同影响喉癌的发生、发展.
