白细胞介素13抗体对支气管哮喘小鼠气道炎症的影响

     

摘要

目的 基于白细胞介素13(IL-13)抗原表位制备抗IL-13抗体,观察其对支气管哮喘小鼠气道炎症的影响.方法 通过软件分析获得IL-13的B细胞表位LTKELIEELSNITQ,合成肽段与KLH蛋白耦联制备多克隆抗体.将支气管哮喘小鼠模型分为IL-13抗体治疗组、兔IgG治疗组和PBS组,同时设正常对照组,观察IL-13抗体对小鼠支气管哮喘的治疗作用.结果 获得针对IL-13全蛋白的多克隆抗体.IL-13抗体组较兔IgG组和PBS组白细胞数量明显减少,支气管肺泡灌洗液中IL-4、IL-5和γ-IFN含量明显降低.结论 IL-13中和抗体对小鼠支气管哮喘症状有明显缓解作用.%Objective To prepare antibody against IL-13 and observe its effects on airway inflammation in bronchial asthmatic mice. Methods The synthesized epitope peptide(LTKELIEELSNITQ),which coupled with KLH,was used as immunogen to generate IL-13 neutralization antibody. A total of 24 BALB/c mice were divided into 4 groups:IL-13 neutralization antibody treatment group,rabit IgG treatment group,PBS treatment group and negative control group. Results IL-13 neutralization antibody was successfully obtained. Compared with rabit IgG treatment group and PBS treatment group,the leucocytes were observably decreased in IL-13 neutralization antibody treatment group, as well as the level of IL-14,IL-5 and γ-IFN. There was no significant difference between rabbit IgG treatment group and PBS treatment group. Conclusion IL-13 neutralization antibody worked effectively on suppressing the airway inflammation in mice bronchial asthmat model.
