Objective To investigate the protective effects of atorvastation on myocardium ischemia reperfusion injury in rats. Methods A total of 54 SD rats were randomized into three groups:atorvastatin group(n=18),ischemia reperfusion group(n=18),and sham-operated group(n=18);the rat model of myocardial ischemia-reperfusion was established,and 8 rats were sacrificed from atorvastatin group and ischemia reperfusion group for Evans blue and TTS staining. Myocardial infarct size of the16 rats were analyzed using image processing software. The level of malondialdehyde (MDA)were measured by thiobarbituric acid method(TBA);tumor necrosis factor-alpha(TNF-α)and myeloperoxidase(MPO)were determined by enzyme-linked immunoadsordent assay(ELISA). 14 days after the operation,the hemodynamic parameters and ventricular remodeling of remain-ing 30 rats were measured. Results Myocardial infarct size of atorvastatin group was lower than ischemia reperfusion group;compared with the sham-operated group,the level of MDA,TNF-αand MPO were increased in both ischemia reperfusion group and atorvastatin group,but rate of increase is lower in atorvastatin group than ischemia reperfusion group;the hemodynamic parameters and ventricular remodeling of rats were significantly im-proved,all the differences were all significant(P<0.05). Conclusion Atorvastatin can reduce the myocardial infarct size,improve the hemody-namic parameters and ventricular remodeling in rat myocardium ischemia-reperfusion injury.%目的:研究阿托伐他汀对大鼠心肌缺血再灌注损伤的保护作用。方法54只SD大鼠随机分为3组:阿托伐他汀组(n=18)、缺血再灌注组(n=18)和假手术组(n=18)。制备大鼠心肌缺血再灌注损伤模型,从阿托伐他汀组和缺血再灌注组各选取8只小鼠进行Evans blue和TTS染色,应用图像分析软件计算梗死的面积,制备大鼠心肌组织匀浆,采用硫代巴比妥酸(TBA)法测定丙二醛(MDA)含量,酶联免疫(ELISA)法测定肿瘤坏死因子α(TNF-α)及髓过氧化物酶(MPO)含量。于术后14 d检测剩余30只大鼠的血流动力学指标及心室重构指标,并对结果进行统计分析。结果阿托伐他汀干预组大鼠与再灌注组比较,心肌梗死面积小于缺血再灌注组,缺血未梗死面积大于缺血再灌注组,心肌梗死面积占缺血总面积的比例小于缺血再灌注组。与假手术组比较,缺血再灌注组及阿托伐他汀组MDA、TNF-α和MPO均增加,但阿托伐他汀组增加较少。血流动力学及左心室功能较缺血再灌注组明显改善,差异有统计学意义(P<0.05)。结论阿托伐他汀可减小心肌缺血再灌注后损伤的梗死面积,改善血流动力学及左心室功能。
