Objective To investigate the effect of Interleukin‐4 on the release of inflammatory factors in primary astrocytes .Methods Primary cultures of astrocytes were isolated from the suckling mouse which was born just for one or two days ,and the astrocyte purity was detected after the third passages culture .Interleukin‐4 was used to stimulate the astrocytes ,and then Q‐PCR was employed to detect the release of various inflammatory factors .Results The purity of the primary astrocytes was up to 95% .The expressions of anti‐inflammatory factors such as ARG1 ,CD206 ,TGF β,IFN β and IL‐10 increased ,while the expressions of pro‐inflammatory cytokines such as TNFα and IL‐6 decreased .Conclusion Interleukin‐4 plays an active role in promoting anti‐inflammatory cytokines and inhibiting pro‐inflammatory cytokines in primary astrocytes .%目的:探讨白细胞介素‐4(IL‐4)对星形胶质细胞炎症因子释放的影响。方法选取新生1~2 d的C57乳鼠,取大脑皮层制备原代星形胶质细胞,传代3次,检测星形胶质细胞纯度,IL‐4刺激后,用Q‐PCR来检测多种炎症因子的释放。结果原代培养的星形胶质细胞纯度高达95%,用IL‐4刺激后,抗炎因子A RG1、CD206、TGFβ、IFNβ、IL‐10表达升高,促炎因子 TNFα、IL‐6表达降低。结论 IL‐4对星形胶质细胞抗炎因子的分泌有促进作用,对促炎因子的分泌有抑制作用。
