假单胞菌属四氯化碳降解菌的筛选和初步鉴定

     

摘要

[目的]为污染地域四氯化碳的生物降解提供一条可行的途径.[方法]用富集培养基对采自四氯化碳污染区域土样中的菌种进行富集培养,然后用筛选培养基对富集培养液中的菌种进行分离和纯化,用气相色谱法测定各样品中的四氯化碳残留量,进而计算出其四氯化碳降解率.[结果]通过试验筛选出了10株具有降解四氯化碳能力的菌种,其编号分别为SL-1、SL-2、SL-3、SL-4、SL-5、SL-6、SL-7、SL-8、SL-9和SL-10,其四氯化碳降解率分别为11.2%、14.5%、14.9%、18.3%、8.8%、20.1%、19.2%、13.2%、17.5%和15.3%,其中菌株SL-6呈灰色,边缘整齐呈半圆形,其四氯化碳降解率最高.生理生化鉴定结果表明,菌株SL-6属于假单胞菌属.[结论]该试验筛选出了10株能降解四氯化碳的菌株,但它们的四氯化碳降解率需要通过诱变育种等微生物技术来提高.%[Objective] The purpose of the study was to provide a feasible approach for the biodegradation of carbon tetrachloride in polluted regions. [Method] The strains in the soil samples collected from the regions polluted with carbon tetrachloride were cultured in enrichment medium by enrichment method, and then the strains in enriched culture medium were isolated and purified with selective medium. The residues of carbon tetrachloride in various samples were determined by gas chromatography, and then their degradation rates of carbon tetrachloride were calculated out. [Result] Ten strains with ability of degrading carbon tetrachloride were screened out through experiments, their serial numbers were SL-1, SL-2, SL-3, SL-4, SL-5, SL-6, SL-7, SL-8, SL-9 and SL-10 resp. and their degradation rates of carbon tetrachloride were 11.2%, 14.5%, 14.9%, 18.3%, 8.8%, 20.1%, 19.2%, 13.2%, 17.5% and 15.3% resp. Among them, the strain SL-6 was gray and had orderly and semicircular edge and its degradation rate of carbon tetrachloride was highest. The results of physiological and biochemical identification showed that the strain SL-6 belong to pseudomonas genus. [Conclusion] Ten strains with ability of degrading carbon tetrachloride were screened out in this experiment, but their degradation rates of carbon tetrachloride needed to be enhanced by microbial techniques including induced breeding.
