血红铆钉菇液体发酵条件的优化

         

摘要

[ Objective] The study aimed to optimize the liquid fermentation conditions of Gomphidius rutilus. [ Method] G. rutilus mycelium was cultured by liquid fermentation, the effects of some influencing factors such as cultivation time and initial pH in medium on the mycelium yield were investigated through the single factor test and the culture conditions suitable to produce the myeelium by liquid fermentation were confirmod through the orthogonal test. [ Result] The single factor test showed that the optimum culture time for G. rutilus mycelium was 6 d at lest, the liquid seed inoculation should be over 10%, the pH value in the medium was as best as 8.5, the liquid volume in flask was 200 mi/S00 mi flask that was beneficial to the growth of mycelium and the optimum rotating speed could be controlled at 180 r/min. The liquid fermentation conditions of G. rutilus optimized by the orthogonal test were as follows: the initial pH value in the medium was 9. 0, the liquid volume in flask was 200 mi/500 ml flask, the liquid seed inoculation was 12.5% , the culture time was 7 d, the culture temperature was 28 ℃, under which, the yield of G. rutilus mycelium could reach to (9. 320 + 0. 151 ) mg/L. [ Conclusion] The study provided the scientific basis for getting high yield of G. rutilus mycelium.%[目的]优化血红铆钉菇菌丝体液体的发酵条件.[方法]采用液体发酵培养血红铆钉菇菌丝体,先用单因素试验分别考察培养时间和培养基起始pH值等因素对菌丝体产量的影响,然后通过正交试验确定适于血红铆钉菇菌丝体液体发酵生产菌丝体的培养条件.[结果]血红铆钉菇菌丝体的最佳培养时间至少为6 d,液体种子接种量应在10%以上,培养基的pH值以8.5为最好,装液量为200mL/500 ml三角瓶对菌丝体的生长较为有利,最适转速控制在180 r/min.通过正交试验优化的血红铆钉菇菌丝体液体发酵条件为:培养基初始pH值为9.0,装液量为200 ml/500 ml三角瓶,液体种子接种量为12.5%,培养时间为7 d,培养温度为28℃.在此条件下,菌丝体产量可迭(9.320±0.151)g/L.[结论]该研究为获得最高产量的血红铆钉菇菌丝体提供了科学依据.
