[目的]对梭梭组织培养的灭菌方法及外植体进行筛选,以期为其组织培养提供指导方法.[方法]以梭梭种子为试验材料,研究不同灭菌剂处理的灭菌效果和不同粒径种子对无菌苗成活率的影响.[结果]梭梭种子最佳灭菌方法为75％酒精5 min +0.1％升汞8min;以直径φ＞2.0mm的种子外植体,组培能获得较高的无菌苗成活率,可达65％以上.[结论]该研究结果建立了适合梭梭种子组织培养的表面灭菌方法.%[Objective] The aim was to select appropriate sterilization methods and explants for tissue culture of Haloxylon ammodendron (C. A. Mey. ). [Method] H. ammudendron ( C. A. Mey. ) seeds were used as experimental materials to investigate the sterilization results of different sterilization treatments and the effect of different seed sizes on the survival rate of aseptic seedlings. [Result] Sterilization of H. ammodendron (C. A. Mey. ) seeds achieved the best result by using 75% alcohol for 15 min and 0.1% mercuric chloride for 8 min. Seeds with diameter (c) >2.0 mm were used as explants and achieved relatively high survival rate of aseptic seedlings, which reached above 65%. [Conclusion] This study established a surface sterilization method for tissue culture of H. ammodendron (C. A. Mey. ) Bunge seeds.
