[Objective] The aim was to investigate the effect of lycium bararum polysaccharides(LBP) on the proliferation of Spermatogonial stem cells(SSCs)cultured in vitro. [Method] Spermatogonial stem cells were isolated and greatly purified from 2 to 5-day-old ICR mouse testes. Spermatogonial stem cells were cultured with LBP at different concentrations in vitro and the quantity of live cells was determined by MTT assay and bromodeoxyuridine (BrdU) -labeledSpermatogonial stem cells. [Result] LBP enhanced the number and percentage of cell survival in a dose dependent manner compared with control, namely serum-free StemPro-34 SFM medium cultured SSCs. The proliferation of Spermatogonial stem cells was significartly promoted by lycium bararum polysaccharide at high concentration( P < 0. 01). [ Conclusion] LBP could promote Spermatogonial stem cells proliferation obviously.%[目的]观察枸杞多糖(LBP)对体外培养的小鼠精原干细胞的增殖作用.[方法]选2~5日龄的ICR小鼠,采用多次贴壁法纯化精原干细胞培养3d,加入不同浓度的LBP,利用MTT比色分析法和Brdu增殖细胞标记法,检测不同浓度LBP对体外培养精原干细胞增殖的影响.[结果]与对照相比,用LBP处理后的精原干细胞的细胞数量明显增多,高浓度的LBP(200 μg/ml)对精原干细胞的增殖有明显的促进作用(P<0.01).[结论]LBP能够促进精原干细胞的增殖.
