硫代巴比妥酸法测定鱼体肌肉中脂质过氧化物含量的方法优化

摘要

[目的] 优化鱼体肌肉组织中脂质过氧化物(LPO)含量的测定条件.[方法] 使用硫代巴比妥酸分光光度法测定LPO含量,设计乙酸用量、十二烷基磺酸钠(SDS)用量、处理时间和温度4因素3水平的正交试验,考察各因素水平组合对鱼体肌肉中LPO含量测定的影响,优化测定条件.[结果]各因素对LPO含量测定结果的影响由大到小依次为SDS用量、乙酸用量、温度、处理时间;通过直观分析和方差分析,得出最优组合为A3B1C3D2 ,即测定的最优组合为20%的乙酸,65 ℃水浴中保持60 min.[结论]该研究可为鱼类氧化应激损伤研究提供方法支持.%[Objective] To improve the optimum condition for measuring the lipid peroxidation (LPO) in the muscles of fish.[Method] The LPO was determined using spectrophotometer with thiobarbituric acid.The orthogonal design with four factors and three levels was conducted to evaluate the level of LPO in the cyprinoid fish.Four factors were included the gradient of acetic acid,sodium dodecyl sulfate,time and temperature,respectively.[Result] The weight sequence was known as SDS,the gradient of acetic acid,temperature,time by comparing the difference of each factor.The results indicated that the optimum group was A3B1C3D2 by visual analysis and variance analys,namely 20% acetic acid,65 ℃ water bath,60 min.[Conclusion] The study can support the research on the demage of oxidative stress in fish.
