趋化因子受体7在人结肠癌组织中的表达及其与临床病理特征的关系

摘要

目的 观察趋化因子受体7 (chemokine receptor 7,CCR7)在结肠癌中的表达情况,探讨CCR7表达水平与临床病理特征的关系.方法 采用Real-time PCR、Western blot及免疫组织化学技术检测CCR7在90例结肠癌组织及相应的癌旁组织中的表达分布情况,并分析其表达差异与结肠癌临床病理特征的相关性.结果 在90例结肠癌组织中,CCR7在结肠癌组织中mRNA表达水平均显著高于癌旁组织(P＜0.05).免疫组织化学检测结果显示,CCR7在结肠癌组织中的蛋白表达定位于细胞质,CCR7蛋白在癌旁组织与结肠癌组织中的阳性表达率分别为36％ (32/90)和77％ (69/90),两者差异有统计学意义(x2=25.689,P=0.000).Western blot结果显示CCR7蛋白在结肠癌组织中的表达水平显著高于癌旁组织.结肠癌组织中CCR7表达水平与结肠癌浸润深度、淋巴结转移、远处转移及TNM分期显著相关(P＜().05),与病人性别、年龄、肿瘤大小及分化程度无明显相关(P＞0.05).结论 CCR7在结肠癌组织中高表达,其表达水平与结肠癌侵袭转移密切相关,有望成为结肠癌治疗新靶点.%Objective To investigate the expression of chemokine receptor CCR7 in colon cancer tissues and its correlation with clinicopathologic parameters of colon cancer.Methods Real-time PCR,Western blotting and immunohistochemistry were employed to detect the CCR7 expression in 90 cases of colon cancer tissues and the adjacent normal tissues.Furthermore,the correlation between the CCR7 expression levels and clinicopathological characteristics of colon cancer was analyzed.Results The CCR7 mRNA level was significantly increased in tumor tissues as compared with that in adjacent normal tissues in 90 cases of colon cancer (P＜0.05).Immunohistochemistry showed that CCR7 protein was located in cytoplasma.In addition,the positive expression rate of CCR7 in tumor-adjacent tissues and colon cancer tissues was 36％ (32/90) and 77％ (69/90) respectively (P＜0.05).Western blotting results showed that the CCR7 protein expression levels were significantly higher in colon cancer tissues than in the adjacent normal tissues.The CCR7 protein expression was tightly correlated with depth of infiltration,lymph nodes metastasis,distant metastasis and TNM stage (P＜0.05),but not with gender,age,tumor size and histopathological differentiation (P ＞ 0.05).Conclusions The CCR7 expression is significantly increased in colon cancer tissues as compared with the adjacent normal tissues.CCR7 is related to the invasion and metastasis of colon cancer,and may be a new target for therapy.
