碱性成纤维细胞生长因子在新生大鼠缺氧缺血性脑损伤后海马神经发生中的作用

         

摘要

Objective To investigate the correlation between basic fibrobtast growth factor (bFGF) and the endogenous repair mechanism of nerve cell after hypoxia-ischemia brain damage (HIBD). Methods A total of 128 SD rats was randomly and equally divided into 4 groups of A(sham operated), B(hypoxia alone),C(ischemia alone) and D(hypoxia plus ischemia). The HIBD model was established, and the positive expression of BrdU and the expression of bFGF mRNA in the hippocampus were detected by immunohistochemistry and RT-PCR, respectively. Results Compared with other groups, the positive cells of BrdU and the expression of bFGF mRNA were signigicantly increased on the 7th and 14th day after HIBD in group D(P<0. 05). Conclusion The nerve cells regenerate in the dentate gyms of the hippocampus after HIBD, and bFGF may take part in the endogenous repair mechanism of nerve cell after HIBD.%目的 探讨碱性成纤维细胞生长因子(bFGF)与缺氧缺血性脑损伤(HIBD)后神经细胞内源性修复机制的相关性.方法 128只SD大鼠随机均分为假手术(A)组、单纯缺氧(B)组、单纯缺血(C)组和缺氧缺血(D)组.建立HIBD模型,免疫组化检测溴化脱氧核糖尿嘧啶(BrdU)阳性细胞表达,RT-PCR检测海马bFGF mRNA表达.结果 与其余各组比较,D组BrdU阳性细胞数及bFGF mRNA表达于HIBD后7、14 d明显增加(P<0.05).结论 HIBD后海马齿状回存在神经细胞再生,bFGF可能参与HIBD后神经细胞内源性修复机制.
