选择液芯胶囊内外菌体密度和胶囊强度为指标,研究接种量、培养批次和钙离子对微囊化乳酸菌高密度培养和胶囊在培养过程中菌体的释放特性的影响。结果表明菌体接种量在5%以下时,随着接种量的增大,相同培养时间内液芯胶囊内的菌体密度增大,且释放量也增大;随着培养批次的增加,液芯胶囊内和培养液中菌体密度都增加,培养至第4批时囊内菌体密度达1.12×1011CFU/g,但继续增加培养批次菌体密度变化不大;培养液中添加碳酸钙可减缓液芯胶囊强度下降,进而延长了胶囊的使用期限。由此确定了乳酸菌高密度培养的最佳条件,即接种量为5%,培养批次为4,MRS培养液中添加0.3%碳酸钙。在此培养条件下,菌体密度达到1.12×1011CFU/g,并能延长胶囊的使用寿命。%The cell density inside and outside liquid-core capsule and capsule intensity were selected as indexes, to evaluate the effects of inoculation amount, culture batches and Ca2+ in MRS medium on the high cell density culture and releasing property of liquid-core encapsuled lactic acid bacteria. The results showed that cell density inside liquid-core capsule increased with increase of the inoculation size less than 5% , and the releasing of cell was also increased. The cell density inside and outside liquid-core capsule also increased with additional culture batches, inside density being 1. 12×1011 CFU/g at the fourth batches. Density changes were not evident with continuously increasing batches. Adding CaCO3 in MRS culture solution slowed down the intensity decline of liquid-core capsule and prolonged its service life. It could be indicated that the optimal high cell density culture condition was 5% inoculation size, four culture batches and 0.3% CaCO3 in MRS culture solution.
