苏姜猪H-FABP基因多态性及其与肉质性状的相关性

             

摘要

本试验探究苏姜猪心脏脂肪酸结合蛋白质( H-FABP)基因的多态性及其与肉质性状的相关性,采用2种内切酶Hinf玉和HaeⅢ的PCR-RFLP分子标记技术,分析了40头苏姜猪H-FABP基因5忆-上游区和第2内含子区的遗传变异情况.结果显示,试验猪群5忆-上游区Hinf玉酶切位点存在多态性,酶切得到HH、Hh、hh 3种基因型,H等位基因频率为0.65,表现为中度多态(PIC=0.3515),第2内含子区Hae Ⅲ酶切位点存在多态性,酶切得到DD、Dd、dd 3种基因型,D等位基因频率为0.70,表现为中度多态(PIC=0.3318);苏姜猪群的Hinf玉位点等位基因频率和基因型频率都处于哈迪温伯格平衡状态(P﹥0.05);H-FABP基因对肌内脂肪含量的影响表现为HH基因型﹥Hh基因型﹥hh基因型及dd基因型﹥Dd基因型﹥DD基因型(P<0.05),对大理石纹的影响表现为HH基因型﹥Hh基因型﹥hh基因型及Dd基因型﹥dd基因型﹥DD基因型(P <0.05).表明苏姜猪H-FABP基因5忆-上游区和第2内含子区的多态性对其肉质性状有重要的影响.%Po1ymorphism of heart fatty acid-binding protein( H-FABP) gene in Sujiang swine and its corre1ation with meat qua1ity were studied. Po1ymerase chain reaction-restriction fragment 1ength po1ymorphysim ( PCR-RFLP ) with two kinds of restriction enzymes of Hinf I and HaeⅢand mo1ecu1ar marker techniques were app1ied to ana1yze the genetic vari-ations of H-FABP gene 5’-upstream region and second intron in 40 Sujiang swine. There was po1ymorphism in 1ocus of the Hinf I in 5’-upstream region with three genotypes of HH, Hh and hh, and the frequency of a11e1e H was 0. 65, showing moderate po1ymorphism(PIC=0. 351 5). In the second intron, po1ymorphism was detected in po1ymorphic 1ocus HaeⅢwith three genotypes of DD, Dd and dd, the frequency of a11e1e D was 0. 70, indicating moderate po1ymorphism(PIC=0. 331 8) as we11. Both Hinf I gene frequencies and genotype frequencies of tested pigs stayed in the Hardy-Weinberg equi-1ibrium ( P﹥0. 05 ) . H-FABP gene significant1y affected the intramuscu1ar fat ( IMF ) content and marb1ing content ( P<0. 05), fo11owing the significance order of HH﹥Hh﹥hh and dd﹥Dd﹥DD for IMF, HH﹥Hh﹥hh and Dd﹥dd ﹥DD for marb1ing content. The resu1ts suggested that the po1y-morphisms in 5’-upstream region and second intron of H-FABP gene had significant effects on meat qua1ity in Su-jiang swine.
