Objective To investigate the characteristics of mpt64 gene mutation of MPT64-negative mycobacterium tuberculosis in Guangdong.Methods 17 MPT64-negative and 186 MPT64-positive clinical isolates of mycobacterium tuberculosis which were stored in Mycobacterium Strain Culture Collection of Guangzhou District were cultured and proliferated.The genes of mpt64 were sequenced and analyzed.Results Mutations were found in 16 MPT64-negative strains;among which,14 got 63-bp deletion at 196-258,1 had a single nucleotide substitution of G to C at 173,and another strain had a nucleotide of A insertion at 353 meanwhile.4 MPT64-positive strains had point substitution mutations,such as C to G at 323,G to A at 127,C to A at 477,and C to T at 654.Conclusion The deletion of 196-258 bases of mpt64 gene may be the molecular mechanism for mycobacterium tuberculosis of non-MPT64 secretory in Guangdong.%目的 了解广东地区MPT64阴性结核分枝杆菌临床分离株的mpt64基因序列特征.方法 对保存于本院“广州地区分枝杆菌菌种保藏库”中的17株MPT64阴性和186株MPT64阳性结核分枝杆菌临床分离株培养增殖后进行mpt64基因序列测定与分析比对.结果 17例MPT64阴性株中有16例存在基因突变,其中14例在196-258位置存在63 bp碱基缺失;1例存在196-258位碱基缺失与173位碱基G→C置换突变;1例存在196-258位碱基缺失与353位碱基A插入突变.186例MPT64阳性株中4例分别存在323位碱基C→G、127位碱基G→A、477位C→A、654位C→T碱基置换突变.结论 mpt64基因196-258位碱基缺失可能是广东地区结核分枝杆菌不表达分泌MPT64分子机制.
