The effects of agitation speed and aeration rate on the growth of Aspergrillus niger WB6002,mannanase activity,dissolved oxygen and residual sugar were investigated in a 7 L batch fermenter.The results revealed that the agitation speed and aeration rate had significant impacts on the growth of A.niger and production of β-mannanase.The highest A.niger biomass (43.98 g/L) and mannanase activity (1117.26 U/mL) were obtained under the optimal operating conditions as follows:agitation speed 900 r/min,aeration rate 1.5 vvm,temperature 35 ℃ and pH 4.5.%采用7L搅拌式发酵罐研究黑曲霉生产β-甘露聚糖酶的扩大培养条件,主要考察了搅拌转速和通气量对黑曲霉WB6002生长、β-甘露聚糖酶酶活、溶氧以及残糖的影响.研究结果表明,搅拌转速和通气量能够显著影响黑曲霉的生长以及β-甘露聚糖酶的合成,并得出了较为优化的培养条件:温度35℃,pH 4.5,搅拌转速900 r/min,通气量1.5 vvm,装液量50%.在此条件下,黑曲霉WB6002的产酶能力最强,甘露聚糖酶的酶活力达到1117.26 U/mL,最大细胞得率达到43.98g/L.
