榨菜下胚轴离体培养植株再生体系的建立

     

摘要

Using the hypocotyl of pickle (Brassica juncea Czern.et Coss. Var. Tumida) material B2-1014 as explant, the effects of hormone concentration and hormone combination on induction of callus, adventitious bud and adventitious root was studied. The results showed that optimum medium for callus, adventitious bud and adventitious root induction was MS+0.5 mg/L 6-BA+0.8 mg/L NAA, MS+2.0 mg/L 6-BA+0.2 mg/L NAA, and MS+0.2 mg/L NAA, respectively; and the induction rate of callus, adventitious bud and adventitious root of B2-1014 could reach up to 90.0%, 92.2%, and 97.8% respectively.%以榨菜(Brassica juncea Czern.et Coss.var.tumida)B2-1014的下胚轴为外植体,研究了不同浓度的激素以及不同激素组合对愈伤组织、不定芽和不定根诱导的影响.结果表明,榨菜茎段离体培养的植株高频再生体系中愈伤组织诱导培养基为MS+0.5 mg/L 6-BA+0.8 mg/L NAA,不定芽诱导培养基为MS+2.0 mg/L 6-BA+0.2 mg/L NAA,不定根诱导培养基为MS+0.2 mg/L NAA,在此培养条件下,B2-1014愈伤组织的诱导率达90.0%,不定芽诱导率达92.2%,不定根诱导率达97.8%.
